摘要
[目的]探讨芍药软肝方对H22肝癌、S180肉瘤荷瘤小鼠的抑瘤作用,并初步探索其作用机制。[方法]采用H22肝癌、S180肉瘤移植瘤小鼠模型,分荷瘤阴性对照组(生理盐水)、芍药软肝方低、中、高剂量组和阳性对照组(环磷酰胺,CTX),观察芍药软肝方对两种荷瘤小鼠肿瘤生长的抑制作用以及芍药软肝方对H22肝癌荷瘤小鼠生存时间的影响;并运用荧光定量RT-PCR法评价芍药软肝方对H22肝癌荷瘤小鼠肿瘤组织cyclinD1表达的作用。[结果]芍药软肝方低、中、高剂量组和CTX组对移植性荷H22肝癌小鼠的抑瘤率分别为17.72%、33.99%、23.73%、43.95%;与生理盐水组相比,芍药软肝方低、中、高剂量组和CTX组瘤重均有显著性差异(P<0.01)。芍药软肝方低、中、高剂量组和CTX组对移植性荷S180肉瘤小鼠的抑制率分别为29.59%、31.34%、11.17%、53.92%;与生理盐水组相比,低剂量组和中剂量组瘤重均有显著性差异(P<0.05)。芍药软肝方各给药组对H22肝癌腹水瘤小鼠均有一定的生命延长作用,中剂量组与生理盐水组生存天数相比(18.42±2.50vs15.75±2.42),具有显著差异(P=0.014)。芍药软肝方能下调H22肝癌小鼠肿瘤组织cyclinD1 mRNA的表达水平,低剂量组或中剂量组与生理盐水组相比有显著差异(P=0.031,P=0.012),高剂量组与生理盐水组相比差异非常显著(P=0.004)。[结论]芍药软肝方对H22肝癌、S180肉瘤荷瘤小鼠有明显的抑瘤作用,其抑瘤作用可能与下调肿瘤组织cyclinD1mRNA表达水平有关。
[Purpose] To investigate the anti-tumor effect and its mechanism of Shaoyao Ruangan Fang in hepatoma H22 and sarcoma S180 bearing mice.[Methods] The models of hepatoma H22 and sarcoma S180 bearing mice were divided into negative control group(normal saline),Shaoyao Ruangan Fang group(low dose,moderate dose,high dose) and positive control group(CTX).Inhibition of Shaoyao Ruangan Fang on tumor in two sorts of tumor bearing mice,and survival of hepatoma H22 bearing mice with Shaoyao Ruangan Fang were investigated.The impact of Shaoyao Ruangan Fang on expression of cyclinD1 mRNA in tumor tissues of hepatoma H22 bearing mice was investigated by quantitative real-time polymerase chain reaction(QRT-PCR).[Results] The inhibitory rate of tumor in hepatoma H22 bearing mice in low,moderate and high dose group and CTX group was 17.72%,33.99%,23.73% and 43.95%,respectively.The weight of tumor in low,moderate and high dose group and CTX group was significantly different with that of saline control group(P〈0.01).The inhibitory rate of tumor in the sarcoma S180 bearing mice in low,moderate and high dose group and CTX group was 29.59%,31.34%,11.17% and 53.92%,respectively.The weight of tumor in low and moderate dose group was significantly different with saline control group(P〈0.05).Shaoyao Ruangan Fang increased the survival of hep atoma H22 bearing mice.There was significant different in survival between mid-dose group and saline control group(18.42±2.50 vs 15.75±2.42,P=0.014).Shaoyao Ruangan Fang down-regulated expression of cyclinD1 mRNA in tumor tissues from hepatoma H22 bearing mice.There was significant difference in positive expression of cyclinD1 between low or moderate dose group and saline control group(P=0.031,P=0.012).Compared with saline control group,the effect of the high dose group was the most prominent and showed significantly different in statistics(P=0.004).[Conclusion]Shaoyao Ruangan Fang has significant antitumor effect in hepatoma H22 and sarcoma S180 bearing mice,with dose-response effect of parabolic type.The possible mechanism of Shaoyao Ruangan Fang inhibiting proliferation was associated with down-regulating expression of cyclinD1 mRNA in tumor tissues.
