摘要
目的:为了对阿米卡星(amikacin)脂质体进行质量评价,测定了阿米卡星脂质体的包封率。方法:采用新型的两步洗脱法,即前25ml 以蒸馏水,后50ml 改为NaCl 溶液作为流动相进行分离。结果:本方法既可保证脂质体在凝胶柱上不被破坏,又可保证脂质体和游离药物在柱上无吸附现象,柱回收率良好。结论:本方法测定阿米卡星脂质体的包封率新颖。
OBJECTIVE:To determine the encapsulation efficiency of liposomal amikacin in order to evaluate the quality of liposomes. METHODS:A novel gel filtration chromatography with two mobile phaseelution, i.e.,the first 25ml distilled water and the following 50ml 0.9 per cent NaCl solution (saline),was used. RESULTS:This method not only protected liposomes from destroying, but also no drug adhesive in gel column. The encapsulation efficiency of liposomal amikacin was 85.3%.CONCLUSION: This analytical method to determine the encapsulation efficiency of liposomal amikacin was novel and reliable.
