分化人脑胶质瘤干细胞的放疗耐受性

摘要 目的:比较不同剂量的放射治疗对人脑胶质瘤干细胞及其分化的胶质瘤细胞的杀灭作用,探讨胶质瘤干细胞与放疗耐受的相关性。方法:分离、培养并纯化人脑胶质瘤干细胞,并同时诱导纯化的肿瘤干细胞向胶质瘤细胞分化,对人脑胶质瘤干细胞及其分化后的胶质瘤细胞分别以0、2、3、5、8、12Gy的6MVX线照射后,继续培养72h,采用MTT法检测细胞的增殖情况。结果:由胶质瘤干细胞分化出的子代胶质瘤细胞对放射治疗的耐受性下降,且随照射剂量增大,细胞生长抑制显著(F=125.971,P<0.001)。结论:脑胶质瘤干细胞分化后,对放射治疗的耐受性下降。 Aim:To compare the different doses of radio ray's therapeutic effects on human brain glioma stem cells (GSC)and glioma cells and explore the relationship between radiotherapy resistance and differential phases of glioma stem cell.Methods:GSC were isolated from glioma,and meanwhile induced into normal differential glioma cells.GSC and differential glioma cells were treated with 0,2,3,5,8,12 Gy 6 MV X ray,and further cultured for 72 h.The proliferation of GSC and differential glioma cells were tested by MTT.Results:The glioma cells derived from GSC were sensitive to radiation therapy,in a dose dependent manner(F=125.971,P0.001).Conclusion:The glioma cells derived from GSC are more sensitive to radiation therapy than GSC.
