摘要 目的:探讨机动车尾气有机提取物的体外遗传毒性作用。方法:用双核细胞微核试验和彗星试验评价机动车尾气有机提取物(2.5、5.0和10.0mg/L)对HepG2细胞的遗传毒性,另设溶剂对照组和阳性对照组,每组6个平行孔。结果:各组双核细胞微核率和Olive尾矩比较,差异有均统计学意义(F=18.608和9.525,P均<0.001)。5.0和10.0mg/L有机提取物组双核细胞微核率高于溶剂对照组,3个剂量组Olive尾矩均高于溶剂对照组(P均<0.05)。结论:机动车尾气有机提取物对HepG2细胞具有明显的遗传毒性。 Aim:To explore the genotoxicity of organic extract of vehicle exhaust on HepG2 cells in vitro.Methods:Cytokinesis-block micronucleus assay and comet assay were used to evaluate the genotoxicity of organic extract of vehicle exhaust(2.5,5.0 and 10.0 mg/L)on HepG2 cells.The solvent control group and positive control group were used as control.Results:There were significant differences in frequency of cytokinesis-block micronucleus and Olive tail moment among five groups(F=18.608 and 9.525,P0.001).A significant increased frequency of cytokinesis-block micronucleus was observed in both 5.0 and 10.0 mg/L dose group compared with the solvent control group(P0.05).A significant increased Olive tail moment was observed in three dose groups compared with the solvent control group(P0.05).Conclusion:The organic extract of vehicle exhaust has a distinct genotoxicity on HepG2 cells in vitro.
