缓释PDGF-BB的纳米珍珠层/聚乳酸/纤维蛋白胶复合支架的细胞相容性研究 被引量:8

Evaluation of Cell Biocompatibility of PDGF-BB Releasing Nano-nacre/PDLLA/FG Composite Scaffold

摘要 目的探讨缓释血小板衍生生长因子BB(PDGF-BB)的纳米珍珠层/聚乳酸/纤维蛋白胶复合支架的细胞相容性。方法将MC3T3-E1细胞分别接种缓释PDGF-BB的纳米珍珠层/聚乳酸/纤维蛋白胶复合支架(实验组)、纳米珍珠层/聚乳酸支架(对照1组)和聚乳酸支架(对照2组),单纯细胞培养为空白对照组。倒置相差显微镜、扫描电镜观察细胞生长情况,MMT法检测细胞增殖功能,碱性磷酸酶(ALP)测定了解细胞成骨分化情况。结果细胞在新型支架上粘附生长良好。各组MTT和ALP值随培养时间延长而增大,各时间点实验组均高于其他三组,差异有统计学意义(P<0.05)。结论缓释PDGF-BB的纳米珍珠层/聚乳酸/纤维蛋白胶复合支架具有良好的细胞相容性,是一种理想的骨组织工程支架材料。 Objective To investigate the cell biocompatibility of a platelet-derived growth factor BB(PDGF-BB) releasing nano-nacre/PDLLA/FG composite scaffold.Methods MC3T3-E1 cells were cultured on PDGF-BB releasing nano-nacre/PDLLA/FG composite experimental group,scaffold,nano-nacre/PDLLA scaffold,PDLLA scaffold and on the empty wells as control group one,control group two and blank-control group,respectively.At the indicated time,cell morphology was observed by inverted phase contrast microscope and scanning electron microscope.In addition,MTT assay and ALP activity test were used to evaluate the cells abilities of proliferation and osteogenetic-differentiation respectively.Results The cells adhered and grew well on the surface of the novel scaffold.At the same time point compared experimental group with other groups,MTT detection and the expression of the ALP in experimental group was the highest at the same time point,there was significant difference(P 0.05).Conclusion The PDGF-BB releasing nano-nacre/PDLLA/FG composite scaffold has good biocompatibility with MC3T3-E1 cells,and it might be a promising biomaterial for bone tissue engineering.
