摘要
目的:比较地塞米松和他克莫司对HaCaT细胞热休克蛋白90(ISP90)表达的影响,并探讨其潜在的临床意义。方法:以含10^(-6)mol/L地塞米松或10^(-6)mol/L他克莫司的DMEM(高糖)培养基培养细胞,在24 h和48 h的不同时相点,采用Real timePCR和Western blotting检测tSP90的表达。结果:地塞米松作用24 h后,HSP90 mRNA和蛋白表达水平均升高,48 h后进一步升高(P<0.05);而他克莫司作用于HaCaT细胞后的不同时相点,tSP90 mRNA和蛋白水平与未处理组相比差异均无统计学意义(P>0.05)。结论:地塞米松可上调角质形成细胞中HSP90的表达,他克莫司不影响角质形成ll胞HSP90的表达。
Objective: To investigate the effects of dexamethasone or tacrolimus on heat shock protein 90 (HSP90) expression in HaCaT cells in vitro. Methods: The expression of HSP90 of the HaCaT cells treated with dexamethasone at 10^-6 mol/L or tacrolimus at 10^-6 mol/L were assessed by RT-real time PCR and Western blotting at settled duration. Results: Comparing with negative control, the expression of HSP90 was significantly increased in the cells after the 24 h and 48 h treatment of dexam- ethasone (P〈 0.05), whereas the effect was not observed in the cells treat with tacrolimus. Conclusions: Dexamethasone could up-regulate the HSP90 expression in HaCaT cells, but tacrolimus did not.
