小干扰垂体特异性转录因子-1对大鼠垂体生长激素肿瘤细胞侵袭行为的影响

摘要 目的:研究垂体特异性转录因子-1(pituitary-specific transcription factor-1,pit-1)siRNA转染大鼠GH3型垂体瘤细胞后对其生物学特性的影响。方法:设计合成pit-1 siRNA,经脂质体(lipofectin)转染大鼠垂体瘤细胞后,通过RT-PCR检测瘤细胞内pit-1 mRNA表达的变化;制备Boyden小室测定pit-1 siRNA对肿瘤细胞侵袭力和趋化作用的影响。结果:pit-1 siRNA转染大鼠GH3细胞24 h后,pit-1 mRNA和pit-1蛋白的表达明显低于阴性对照组(P<0.05)。透过基质凝胶多孔滤膜的大鼠GH3细胞瘤细胞的数量都显著低于阴性对照组(P<0.05)。结论:pit-1对大鼠垂体瘤细胞的侵袭性呈正相关,在生长激素腺瘤的发病机制中起重要作用。 Objective:To study the effect of the biological characteristics with the pituitary-specific transcription factor-1 siRNA-transfected rat GH3 Pituitary tumor cells.Methods:Designed synthetic siRNA,liposome(lipofectin) transfected rat pituitary tumor cells by RT-PCR,assayed inhibition of tumor cell pit-1 mRNA and pit-1 protein expression results,preparation of Boyden chamber determination of pit-1siRNA on tumor cell invasion and chemotaxis effects.Results:Pit-1 siRNA-transfected rat GH3 cells after 24 h,pit-1 mRNA expression was significantly lower than the negative control group(P〈0.05).Through the porous membrane matrix gel in rat GH3 cells the number of tumor cells were significantly lower than the negative control group(P〈0.05).Conclusion:Pit-1 in rat Pituitary adenoma cells(GH3) is positively correlated with aggressive,play an important role with the growth hormone in GH3 Pituitary adenoma cells in the pathogenesis.
