ImmuKnow^(TM)免疫细胞功能检测在肝移植术后排斥反应中的意义 被引量:1

Significance of adenosine triphosphate content in CD_4^+ T lymphocytes assay to the rejection after liver transplantation

摘要 目的探讨CD4+T淋巴细胞内ATP检测在肝移植术后排斥反应中的作用及意义。方法选取肝移植术后患者101例,共采集外周血样本142例次。标本检测采用ImmuKnowTM免疫细胞检测试剂盒进行。根据患者是否发生排斥反应及不同的免疫状态,分析CD4+T淋巴细胞内ATP与肝移植术后排斥反应的关系。结果本组患者共发生排斥反应5例次,排斥反应组与非排斥反应组的CD4+T淋巴细胞ATP值分别为(762.00±135.34)、(390.23±238.77)ng/ml,P=0.001。低免疫应答组、正常免疫应答组和高免疫应答组肝移植术后排斥反应发生率分别为0、1.7%(1/59)和9.3%(4/43),P=0.044。他克莫司(FK506)低浓度组和高浓度组CD4+T淋巴细胞ATP值比较无统计学差异,FK506浓度与CD4+T淋巴细胞ATP无直线相关关系(r=-0.055,P=0.518)。结论外周血CD4+T淋巴细胞ATP对肝移植术后排斥反应的防治有重要意义。 Objective To explore the function and significance of adenosine triphosphate (ATP) content in CD4^+ T lymphocytes to reject after liver transplantation (LT). Methods This study contained 101 patients post-LT and 142 peripheral blood samples collected from the patients. ImmuKnow test kits for immune cell function were used to assay the blood samples. According to the rejection and different immune states, the relationship between ATP values and rejection post-LT was analyzed. Results In our study, there were five examples of rejection totally. The average ATP value in rejection group and non-rejection group was (762.00 ±135.34 ) ng/ml and ( 390.23 ±238.77 ) ng/ml respectively ( P = 0.001 ). According to the ATP level, the patients were divided into low immune response group ( 〈 225 ng/ml) , normal immune response group (225-525 ng/ml) and high immune response group ( 〉 525 ng/ml) with the rejective rate of 0, 1.7% (1/59) and 9.3 % (4/43) respectively (P = 0. 044). The patients were divided into low concentration group and high concentration group based on the concentrations of tacrolimus. There was no linear correlation between the concentrations of taerolimus and CD4^+ T cell ATP values (P = 0.518 ). Conclusion The ATP content in CD4^+ T lymphocytes assay is significant to the rejection post-LT.
