摘要
目的 :建立用毛细管区带电泳快速测定血清中氯氮平含量的方法。方法 :采用 33.5mmol·L-1磷酸盐缓冲液 (pH6 .47) ,用紫外检测器在 2 5 4nm波长处检测 ,以外标法定量。结果 :线性范围 0 .1~ 10 μg·ml-1,最低检测限 0 .1μg·ml-1。平均回收率 ( 97.47± 2 .8) %。日内与日间RSD分别为 2 .6 3%和 3.81%。结论 :毛细血管带电泳法是快速测定人血清中氯氮平含量的较好方法。
OBJECTIVE:To establish a method for rapid determination of clozapine lever in serum with capillary zone electrophoresis.METHODS: 33.5 mmol·L -1 phosphate buffer solution (pH 6.47 ) was used,UV detector at 254 nm,clozapine leve was determined by means of external standard method.RESULTS:The linear range was 0.1 ~10 μg·ml -1 ( r = 0.997 ),detection limit 0.1 μg·ml -1 ,the average recovery ( 97.4 ± 2.8 )% with RSD 2.63% and 3.81% for within day and day to day test respectively.CONCLUSIONS:Determination of clozapine level in serum by capillary zone electrophoresis is a good method.
