山楂叶总黄酮对非酒精性脂肪性肝炎大鼠肝脏PPARs表达的影响 被引量:9

Effect of TFHL on the expressions of PPARs in the livers of rats with nonalcoho1ic steatohepatitis

摘要 目的:观察山楂叶总黄酮(TFHL)对高脂饮食诱导的非酒精性脂肪性肝炎(NASH)大鼠肝组织PPARα、PPARγ表达的影响,探讨TFHL防治NASH的作用机制。方法:以高脂饮食喂养12周建立NASH大鼠模型,分别以250、125mg.kg-1.d-1的TFHL和195.4mg.kg-1.d-1的多烯磷脂酰胆碱进行干预,RT-PCR检测肝组织PPARα、PPARγmRNA的表达,ELISA法检测大鼠肝组织匀浆TNF-α、PPARα的含量。结果:NASH模型组大鼠肝组织PPARαmRNA和蛋白、PPARγmRNA表达均较正常组明显降低,肝组织匀浆TNF-α含量较正常组显著增高,PPAR-α、γ基因mRNA表达均与TNF-α水平呈明显负相关。TFHL高、低剂量组和多烯磷脂酰胆碱组大鼠肝组织匀浆TNF-α含量均较模型组大鼠显著降低;肝组织PPARα基因mRNA和蛋白、PPARγmRNA表达均较模型组明显增高。结论:PPARα、PPARγ参与了非酒精性脂肪性肝病的损伤过程,可能是NASH发生的重要机制,TFHL和烯磷脂酰胆碱可能通过促进PPARα、PPARγ的表达,降低TNF-α的水平,从而减轻肝脏的炎性病变,有效地防治NASH。 Objective: To observe the expressions of PPARα,PPARγ in the livers of rats with Nonalcoho1ic Steatohepatitis(NASH) induced by high-fat diet,and on which the TFHL affects.Methods: The model rats of NASH had been induced by 12 weeks of high-fat diet,simultaneously,the model rats were treated with TFHL according to 250,125mg·kg-1·d-1,and with Essentiale according to 195.4mg·kg-1·d-1,the mRNA expressions of PPARα,PPARγ in the rat livers were detected with RT-PCR,the amounts of TNF-α,PPARα in the serum and livers of rats were detected with ELISA.Results: The expressions of the protein and mRNA of PPARα,and the expressions of PPARγ mRNA in the livers of model rats were obviously lowered than the normal,the amount of liver TNF-α was enhanced the normal.The expressions of PPARα mRNA showed negative correlation with the level of liver TNF-α.The inflammatory activity of rat liver and amount of TNF-α in rat liver in the TFHL high and low doses groups and Essentiale group were obviously lowered than the model.The expression of TNF-α gene mRNA,the amounts of serum and liver Leptin showed no difference.The expressions of PPARα protein and mRNA,and PPARγ mRNA in the livers of model rats were obviously enhanced than the model.Conclusion: PPARα and PPARγ take part in the injury process of Nonalcoho1ic fatty liver disease,maybe which is the principal mechanism of NASH onset.TFHL and Essentiale can protect liver and effectively prevent and treat NASH due to promoting the expressions of PPARα and PPARγ,and lowering the level of TNF-α.
