大菱鲆IDH-1基因型分析及其表型与生物学性状的相关性研究

摘要 [目的]研究大菱鲆IDH-1(异柠檬酸脱氢酶-基因座位1)同工酶表型与生物学性状的关系。[方法]采用同工酶水平切片淀粉凝胶电泳及特异性染色方法,研究了60尾人工养殖大菱鲆IDH-1的表达,并对其IDH-1表型以及生物学指标进行了检测,分析IDH-1同工酶表型与生物学指标的关系。[结果]在大菱鲆同工酶电泳中得到2种基因型A1和A2,其中基因型A1 53尾,基因型A2 7尾。在116日龄和165日龄的大菱鲆群体中,基因型为A2的大菱鲆个体的体重、体长和体高的平均值均高于A1个体。A2基因型的116日龄个体的体高、165日龄个体的体重和体高,均明显高于A1基因型,差异极显著(P<0.01)。[结论]该研究为进一步研究大菱鲆的标记辅助育种提供了参考。 [ Objective] The research aimed to study the correlation between the isozyme phenotype of isocitrate dehydrogenase-1 (IDH-1)and the biological characters of Scophthalmus maximus L.. [ Method] Using isozyme starch gel electrophoresis and specific dying method,the expres- sion of IDH-1 in 60 S. maximus artificially cultured was studied. The phenotype of IDH-1 and biological indices of S. maximus were detected and the correlation between the isozyme phenotype and the biological indices was analyzed, [ Result ] 2 genotypes of IDH-I ( At and A2 ) of S. maximus were obtained by using the isozyme electrophoresis and 53 individuals showed genotype A~ and 7 individuals showed genotype As. In the population of 116-day-old and 165-day-old S. maximus, the average values of body'weight, body length and body height of individuals with geno- type As were all higher than that with genotype AI. The body height of ll6-day-old S. maximus with genotype As and the body weight and body height of 165-day-old S. maxim~ with genotype As were obviously higher than that with genotype A~ ,with extremely significant difference (P 〈 0.01 ). [ Conclusion] The research provided references for further studies on the marker-assisted breeding of S. ma^imus.
