摘要
【目的】观察耳针对糖尿病大鼠胰腺的保护作用。【方法】选用SPF级SD大鼠40只,正常组10只,另30只大鼠采用腹腔注射链脲佐菌素(剂量55 mg/kg)法复制糖尿病大鼠模型,造模成功后随机分3组:模型组、二甲双胍组(剂量52mg.kg-1.d-1)、耳针组。治疗前,各组大鼠测空腹血糖、糖化血红蛋白,治疗30 d后再检测各组大鼠血糖、糖化血红蛋白、血胰岛素、C肽水平,并在光镜及电镜下观察大鼠胰腺形态学改变。【结果】各组空腹血糖、糖化血红蛋白水平治疗前与正常组比较均显著升高(P<0.01),治疗30 d后,模型组较正常组显著升高(P<0.01),二甲双胍组、耳针组与模型组及治疗前比较显著下降(P<0.05)。治疗后二甲双胍组和耳针组大鼠胰岛素和C肽水平均有不同程度升高,其中二甲双胍组与模型组比较差异有统计学意义(P<0.05)。大鼠胰岛组织光镜和电镜下形态结构的观察结果表明:模型组大鼠受损伤程度尤为明显,而耳针组和二甲双胍组大鼠胰岛组织结构损伤较轻。【结论】耳针除了具有改善糖代谢的作用外,还可以一定程度促进胰岛素和C肽的释放,保护糖尿病大鼠的胰腺组织。
Objective To observe the protective effect of ear acupuncture on the pancreas of diabetic rats.Methods Forty SD rats with specific-pathogen free were used for the experiment.Of them,10 served as the normal control,the other 30 rats were given intraperitoneal injection of streptozotocin(STZ,55 mg/kg) to induce diabetes.After modeling,the diabetic rats were randomized into model group,metformin group(52 mg·kg-1·d-1)and ear acupuncture group.Before treatment we measured rat fasting blood glucose(FBG) and glycated hemoglobin in each group.After treatment for 30 days,we measured FBG,glycated hemoglobin,blood insulin,and C peptide.Under optical microscope and electron microscope,we also observed pancreatic morphological changes of the rats.Results Before treatment,FBG and glycated hemoglobin in diabetic rats were significantly increased as compared with the normal group(P〈0.01).After treatment for 30 days,FBG and glycated hemoglobin were significantly decreased in model group(P〈0.01 compared with those in the normal group),metformin group and ear acupuncture group(P〈0.05 compared with those before treatment).Blood insulin and C peptide were increased to various degrees in metformin group and ear acupuncture group,and the difference was significant between metformin group and model group(P〈0.05).The results of pancreatic morphological observation under optical microscope and electron microscope showed that the pancreatic injury was obvious in the model group,and was mild in metformin group and ear acupuncture group.Conclusion Ear acupuncture has an effect on protecting pancreatic tissue by improving sugar metabolism and by promoting the release of insulin and C peptide.
