摘要
[目的]探讨研磨法和浸提法测定菜心叶片叶绿素含量的优劣及快速测定的步骤。[方法]以菜心为试材,浸提法设5个处理:95%乙醇、80%丙酮、丙酮∶乙醇=1∶1、丙酮∶乙醇=1∶2和丙酮∶乙醇=2∶1,Arnon研磨法作为对照,研究不同提取液对菜心叶片叶绿素提取效果和叶绿素稳定性的影响。[结果]浸提法要优于Arnon研磨法;浸提法中,混合液浸提法的提取率和提取液的稳定性都优于单一溶剂浸提法,其中菜心叶绿素提取效果最好的是丙酮与乙醇体积比为2∶1,单一的丙酮或乙醇浸提法效果较差。[结论]丙酮与乙醇体积比为2∶1的混合液浸提法直接浸提菜心叶片叶绿素的优点是步骤简单,操作方便,节省时间,稳定性好,可以快速测定大批量样品的叶绿素含量。
[Objective] The study aimed to explore the advantage and disadvantages for the determination of the chlorophyll content in the leaves of flowering Chinese cabbage by grinding method and leaching method and the steps of rapid determination.[Method] With the flowering Chinese cabbage as the tested material,the leaching method was set up 5 treatments as 95% ethanol,80% acetone,acetone∶ethanol = 1∶1,acetone∶ethanol = 1∶2 and acetone: ethanol = 2∶1,Arnon grinding method as the CK,effects of the different extracts on the extraction effect of and stability in the leaves of the flowering Chinese cabbage.[Result] The leaching method was better than the grinding method.As for the leaching method,the extraction rate by the mixture liquid leaching method and the stability of mixture extracting liquid were better than that by a single solvent leaching method,in which,the leaching method including the mixture liquid of the acetone and ethanol with the volume rate of 2∶1 could get the best extraction effect on the chlorophyll in the flowering Chinese cabbage and the single acetone or ethanol got the worst extraction effects.[Conclusion] The mixture liquid of the acetone and ethanol with the volume rate of 2∶1 in directly extracting the chlorophyll in the flowering Chinese cabbage had the advantages of simple steps,convenient operation,good stability and saving time,which could rapidly determine the chlorophyll content in the multitudinous samples.
