摘要
通过人工授粉获得铁皮石斛蒴果,再以蒴果种子为材料进行组培快繁试验,结果表明:人工杂交育种结实率达83%。经种子萌发诱导培养、原球茎增殖培养和壮苗生根培养等试验的结果表明:种子萌发最佳培养基为1/2MS+蛋白胨2 g/L,发芽率达到95%;原球茎增殖最佳培养基为1/2MS+6-BA 1.0 mg/L+NAA 0.5 mg/L+15%马铃薯,增殖系数达到5.7;壮苗生根最佳培养基为1/2MS+NAA 0.5 mg/L+15%香蕉,苗高7.2 cm、根数5.6条;组培苗移栽成活率达90%以上。
An experiment of rapid tissue-culture propagation on Dendrobium candidum seeds were conducted, that capsule have been gained through artificial pollination. The results showed that the seed setting rate of artificial hybrid breeding was 83%. The test results from induction of seed germination, rapid propagation of protocorm and rooting culture for strong sprouts showed that the best medium of seeding germination, protocorm rapid propagation and strong sprout rooting culture were I/2MS + peptone 2g/L, 1/2MS +6 - BA 1.0 mg/L + NAA O. 5 mg/L + 15% potato and 1/2MS + NAA 0. 5 mg/L + 15% banana respectively, with the germination rate of 95%, multiplication coefficient of 5.7 and seedling height of 7.2cm and root number of 5.6 piece, respectively, the transplantation survival rate were over 90%.
