氢氧化锌与低分子量透明质酸复合佐剂对甲肝乙肝联合抗原的体液免疫增强作用 被引量:9

Enhancement of humoral immunity of hepatitis A and B combined antigen by zinc hydroxide and low molecular weight hyaluronic acid compound adjuvant

摘要 目的探讨氢氧化锌与低分子量透明质酸(Low molecular weight hyaluronic acid,LHA)复合佐剂对甲肝乙肝联合抗原诱导小鼠体液免疫应答的影响。方法在甲肝乙肝联合抗原中加入不同配比的Zn(OH)2与LHA复合佐剂,经皮下免疫ICR小鼠,并设铝佐剂组、生理盐水对照组和单纯抗原组。分别于免疫后4、8、12和16周采血,分离血清,ELISA法检测小鼠血清抗-HAV IgG和抗-HBsAg IgG水平。结果除对照组小鼠血清检测不到抗-HAV IgG抗体外,其余各组小鼠血清抗-HAV IgG水平在12周时达峰值,抗-HBsAg IgG水平在8周时达峰值,以后逐渐下降;免疫后4、8、12和16周,多数复合佐剂组抗-HAV IgG和抗-HBsAg IgG水平与抗原组和铝佐剂组相比,显著增强(P<0.05),且抗体水平持续时间较长。结论适当剂量配比的氢氧化锌与LHA复合佐剂能增强甲肝乙肝联合抗原诱导的体液免疫应答。 Objective To investigate the effect of zinc hydroxide[Zn(OH)2] and low molecular weight hyaluronic acid (LHA) compound adjuvant on humoral immunity induced by hepatitis A and B combined antigen in mice. Methods Zn (OH)2/LHA compound adjuvant with various formula were added into hepatitis A and B combined antigen respectively. ICR mice were immunized s. c. with the mixture, using aluminium hydroxide adjuvant, physiological saline and adjuvant-free antigen as control. Serum samples were collected 4, 8, 12 and 16 weeks after immunization and determined for anti-HAV IgG and anti-HBsAg IgG levels by ELISA. Results Except those in physiological saline control group, the serum anti-HAV IgG and anti-HBsAg IgG levels of mice in various groups reached peak values 12 and 8 weeks after immunization respectively then decreased gradually. Most of the anti-HAV IgG and anti-HBsAg IgG levels of mice 4, 8, 12 and 16 weeks after immunization with hepatitis A and B combined antigen containing Zn(OH)J LHA compound adjuvant, at various dosages, increased significantly as compared with those in aluminium hydroxide adjuvant and adjuvant-free antigen control groups (P 〈 0. 05), and lasted for a long time. Conclusion The Zn(OH)2/LHA compound adjuvant with appropriate formula enhanced the humoral immune response induced by hepatitis A and B combined antigen.
