摘要
目的:观察养阴清肺汤对博莱霉素(bleomycin,BLM)致大鼠肺纤维化模型的干预作用和转化生长因子-β1(transforminggrowth factorβ1,TGF-β1)的影响,探讨养阴清肺汤参与肺纤维化的机制。方法:健康SD大鼠60只随机分3组:正常对照组(生理盐水),模型组(博莱霉素),治疗组(博莱霉素+养阴清肺汤)。分别在第71,42,1,28天,每组5大鼠,HE染色观察支气管肺组织病理改变,免疫组化观察肺组织中TGF-β1表达。结果:(1)治疗组大鼠肺泡炎和肺纤维化发生率明显降低,且程度轻,无严重肺纤维化发生。(2)治疗组肺间质中TGF-β1表达比模型组明显减少(P<0.05)。结论:养阴清肺汤能减轻博莱霉素诱导的大鼠肺纤维化,可能是通过下调TGF-β1的表达而起作用。
Objective: To study the effect of Yangyinqingfei decoction inhibitor on bleomycin-induced pulmonary fibrosis in rats and its possible mechanism.Methods: Sixty female were randomly divided into three groups: the model group,the control group and Yangyinqingfei decoction inhibitor treatment group,in which the rats were intratracheally instilled with bleomycin.From the first day after bleomycin challenge,by every day,treatment group was given daily Yangyinqingfei decoction,the other group were given sterile saline.Five rats were killed each group respectively at the time of the seventh day,the fourteenth day,the twenty-first day and the twenty-eighth day.The pathological changes of lung tissue by hematoxylin-eosin staining were investigated.The protein expression of TGF-β1 in bronchial lung tisusue were examined by immuohistochemical analysis.Results: Alveolitis in the model group and treatment group were significantly lower as compared with the control,level of pulmonary fibrosis as inhibited evidently after the treatment by Yangyinqingfei decoction inhibitor.The protein expression level of TGF-β1as inhibited evidently after the treatment by Yangyinqingfei decoction inhibitor.conclusion: Yangyinqingfei decoction inhibitor can alleviated BLM induced pulmonary fibrosis in rat,inhibiting the protein expression of TGF-β1 may be one of the mechanisms.
