摘要
目的:筛选九节龙的抗炎止咳化学组分并对其进行含量测定。方法:采用高效液相色谱法测定九节龙中5种抗炎止咳化学组分的含量。测定没食子酸、岩白菜素的色谱条件参照《中国药典》执行。测定杨梅素、山柰酚、槲皮素的色谱柱为Waters Symmetry-C18(250mm×4.6mm,5μm),流动相为甲醇-0.4%磷酸(45:55),流速为0.8mL·min-1,柱温为30℃,检测波长为368nm。结果:没食子酸、岩白菜素、杨梅素、山柰酚、槲皮素分离度良好,标准曲线在检测范围内均呈良好线性关系;平均加样回收率均>97%,RSD均<2.0%(n=6)。结论:九节龙的叶中5种组分的含量均明显高于全株和茎,采收时间以果期为佳。该测定方法准确、稳定、重复性好,可为九节龙药材质量标准的制定奠定基础。
OBJECTIVE:To screen and determine the chemical components for anti-inflammatory and relieving cough of Ardisia pusilla.METHODS:HPLC method was adopted to determine the contents of 5 chemical components for anti-inflammatory and relieving cough of A.pusilla.The content determination of gallic acid and bergeninum were conducted according to chromatogram conditions stated in Chinese Pharmacopeia.The determination of myricetin,kaempferol and quercetin were performed on Waters Symmetry-C18(250 mm×4.6 mm,5 μm) column with mobile phase consisted of methanol-0.4% phosphoric acid(45:55) at a flow rate of 0.8 mL·min-1.The column temperature was 30 ℃ and detection wavelength was set at 368 nm.RESULTS:The chemical compounds of gallic acid,bergeninum,myricetin,kaempferol and quercetin had a high resolution.The standard curves of the compounds had good linearity in the detection ranges.Average recoveries were all higher than 97% and the RSDs were all lower than 2.0%(n=6).CONCLUSION:The contents of 5 compounds in the leaves are obviously higher than that in the whole plant and the stem.The best harvesting time is fruit period.This method is accurate,stable and reproducible,and it can lay a foundation for the establishment of quality standard for A.pusilla.
