摘要
[目的]优化ZTCl+1天然澄清剂对降糖甲片水提取液的澄清工艺.[方法]以多糖保留量和梓醇保留量为评价指标,采用正交设计试验优化澄清工艺,并将结果与醇沉工艺结果进行比较.[结果]最佳澄清工艺为药材水提取液浓缩至药材(g)与水提取浓缩液(mL)之比为1∶7,在60℃水浴先加入0.30mL/g的1%(质量分数)A组分溶液,后加入0.15mL/g的1%(质量分数)B组分溶液.按照此工艺操作,得多糖和梓醇保留量分别是67.1,0.91g/kg,明显高于醇沉法的34.8,0.71g/kg.[结论]与醇沉工艺比较,该澄清工艺更简单,成本低,有效,更能保留有效成分,如多糖与梓醇.
OBJECTIVE To optimize the clarifying technology of ZTCl+1 natural clarificant for water extract of the JiangtangjiaPian.METHODS The evaluation index was the remained quantities of polysaccharide and catalpol,and the clarifying technology was optimized by orthogonal design test and its result was compared with the result of alcohol sedimentation technology.RESULTS The optimum clarifying technology was that extract solution of Chinese crude drugs was condensed into Chinese crude drugs(g) versus the extract solution(mL) 1∶7,and it was added at 60 ℃of water bath that first 1%(mass fraction) of 0.30 mL/g of component A(ZTC1+1 clarificant) and after 1%(mass fraction) of 0.15 mL/g of component B(ZTC1+1 clarificant).The remained contents of polysaccharide and catalpol were 67.1 g/kg and 0.91 g/kg by ZTCl+1 natural clarificant and 34.8 g/kg and 0.71 g/kg by alcohol sedimentation technology,respectively.CONCLUSION As compared with alcohol sedimentation technology,the advantage of the clarifying technology of ZTC1+1 natural clarificant is more simple operation,lower cost and better clarification with less loss of effective ingredients such as polysaccharides and catalpol.
