摘要 [目的]研究氟苯尼考在日本鳗鲡和欧洲鳗鲡体内的药代动力学特征。[方法]氟苯尼考以混饲口灌方式给药,剂量为30 mg/kg,药时数据利用DAS软件进行药动学分析。[结果]日本鳗鲡和欧洲鳗鲡体内药动学的房室参数:吸收速率常数(Ka)分别为0.329和0.449 l/h,消除相半衰期(t1/2β)为44.266和12.690 h。非房室参数:药时曲线下面积(AUC0-∞)分别为257.099和285.945 mg.h/L,平均滞留时间(MRT0-∞)为18.370和14.227 h,半衰期(t1/2)为12.341和9.919 h,达峰浓度(Cmax)为15.92和20.39μg/ml,达峰时间(Tmax)均为4 h。[结论]建议在鳗鲡中使用氟苯尼考进行治疗时可采用30 mg/kg体重的剂量,给药间隔为12 h,即每天2次。 [ Objective ] To study the pharmacokinetic of florfenicol in A. japonica and A. Anguilla. [ Method ] 30mg/kg florfenicol was orally administrated, and the pharmacokinetics was analyzed using DAS software. [ Result ] Pharmaeokinetics parameters were figured out using a two-compartmental and non-compartmental model. In A. japonica and A. Anguilla, absorption rate constant (Ka) were 0. 329 and 0. 449 l/h; half-life of drug during elimination phase ( t1/2β ) were 44.266 and 12. 690 h ; area under the concentration-time curve (AUC0-∞ ) were 257. 099 and 285. 945 mg · h/L; mean residence time (MRT0-∞ ) were 18. 370 and 14.227 h; half-life of drug (t1/2 ) were 12. 341 and 9. 919 h; maximum concentrations ( Cmax ) were 15.92 and 20.39 μg/ml, respectively; and time to maximum concentration ( Tmax ) were both 4 h. [ Conclusion] It was suggested that twice dose a day of 30 mg/kg body weight would be acceptable for treatment of eel with bacterial infections.
