摘要
目的观察高糖环境下豚鼠膀胱Cajal间质细胞(interstitial cells of Cajal,ICCs)内钙离子荧光及其超微结构的改变,探讨糖尿病膀胱(DCP)的发病机制。方法采用原代培养,激光共聚焦、透射电镜观察5、10、15mmol/L葡萄糖浓度培养24、72h的ICCs,扫描记录胞内荧光强度值,观察其超微结构。结果糖浓度增高培养时间延长,ICCs内钙离子荧光值升高,胞内线粒体大小不等、数量减少、空泡样变甚至溶解,胞质广泛溶解,突起消失。结论高糖环境对豚鼠膀胱ICCs电生理学、超微结构造成显著影响,这种改变可能是造成DCP发病的重要原因之一。
Objective To observe a high glucose environment in guinea pig bladder of interstitial cells of Cajal(interstitial cells of Cajal,ICCs) Ca2+ fluorescence changes of diabetic bladder(diabetic,cystopathy,DCP) in the pathogenesis and ultrastructural.Methods Primary culture ICCs bladder cells observe ICCs at different concentrations(5,10,15 mol/L) and different times(24,72 h),by confocal laser scanning technology and transmission electron microscope,get the calcium fluorescence titer and their ultrastructural of ICCs.Results With the of increased glucose concentration,the duration of culture,ICCs Ca2+ fluorescence values increased.Their mitochondrion were inequality of size,abated of quantity,cavity affected and even dissolution.Cytoplasm were deliquesced generally,scabrosity vanished.Conclusion High glucose environment on guinea pig bladder ICCs electrophysiology and ultrastructural have a significant impact,such alterations may be one of the important reasons causing the onset of DCP.
