摘要
目的探讨何首乌饮对衰老大鼠下颌下腺神经生长因子(NGF)、表皮生长因子(EGF)基因表达的影响。方法选用8周龄SD雌性大鼠90只,用D-半乳糖制造衰老大鼠模型,何首乌饮的干预作用分治疗和预防两部分,用原位杂交法检测各组大鼠下颌下腺NGF mRNA、EGFmRNA表达的情况。结果预防性实验部分的组间NGF mRNA、EGF mRNA表达比较差异有统计学意义(P<0.01),其中以正常组最高,模型组最低,各预防组处于中间水平,其中预防中剂量组最高。治疗性实验部分的组间NGF mRNA、EGF mRNA表达比较差异有统计学意义(P<0.01),其中以阴性对照组最高,自然恢复组最低,各治疗组处于中间水平,其中中剂量组最高。结论何首乌饮可在基因转录水平提高NGF、EGF的表达。
Objective To observe the effects of Heshouwuyin on the expressions of NGF mRNA and EGF mRNA in submandibular glands of aging rats.Methods D-galactose senescence model was adopted.90 female SD rats aged eight weeks were randomly divided into preventive and treatment experimental groups.In-situ hybridization method was used to observe the expression changes of EGF mRNA and NGF mRNA in submandibular glandular cells of every group.Results In preventive groups,both NGF mRNA and EGF mRNA expressions in submandibular gland cells were significantly statistic different compared with those of normal or model group(P〈0.01).Among preventive,normal and model groups,NGF mRNA and EGF mRNA expressions of normal group were the highest,those of model rats were the lowest,those of middle dose of preventive rats were better than those of the others.In treating groups, both NGF mRNA and EGF mRNA expressions of submandibular gland cells had significant differences in comparison with those of normal control group or S-R group (P〈0.01).The effect of middle dose of treatment rats was better than that of the others.Conclusions Heshouwuyin could increase the expressions of NGF and EGF by promoting the gene transcription process.
