理冲汤对子宫肌瘤模型大鼠血清激素水平及组织病理改变的影响 被引量:10

摘要 目的观察理冲汤对子宫肌瘤模型大鼠血清激素水平和病理改变的影响,探讨理冲汤治疗子宫肌瘤的可能机制。方法采用雌、孕激素共同注射的方法建立大鼠子宫肌瘤模型。将大鼠分为正常对照组、模型对照组、理冲汤大剂量组,理冲汤小剂组、两药对照组。各组灌胃给药4周后心脏取血,采用放射免疫法检测血清性激素水平,HE染色镜下观察大鼠子宫平滑肌病理改变。结果模型对照组大鼠血清E2、P、LH水平较正常对照组显著升高;灌胃给药后,理冲汤大剂量组、理冲汤小剂量组E2、P、LH水平明显降低;但理冲汤对血清FSH水平的影响各组间比较无显著性差异。光学显微镜下观察模型对照组大鼠子宫平滑肌增生、肌层增厚,肌细胞排列紊乱。理冲汤大剂组平滑肌层较厚,肌细胞排列整齐,胞核呈长梭形或杆状,核仁不清。理冲汤小剂组平滑肌层较薄,肌细胞排列整齐,胞核呈长梭形或杆状,核仁不清。结论理冲汤能明显降低E2、P、LH水平,并能改善子宫平滑肌的病理状态,提示理冲汤治疗子宫肌瘤的作用机制可能与调节内分泌、改善子宫平滑肌的病理状态有关。 Objective It is to observe the effect of Lichong decoction on serum sex hormone level and the pathologic changes of uterine tissue in rats with hysteromyoma,and to explore its possible mechanism of the treatment for this disease.Methods Uterine leiomyoma models of rats were established by intramuscular injection of estrogen and progesterone.Then the rats were divided into normal group,model group,high and low dosage of Lichong decoction groups,two drug control group.After 4 weeks' treatment by intragastric administration,blood was gotten from heart in every group,and the level of serum sex hormone was detected by radio-immuno-detection,and the pathological changes of uterine smooth muscle of the rats were observed under microscope after HE stain.Results Compared with those in normal control group,serum sex hormone levels such as E2,P,LH in model group were increased significantly.After treatment by intragastric administration,these levels were obviously decreased in high and low dosage of Lichong decoction groups,but Lichong decoction had no significant effect on serum FSH level in each group.Light microscope observation showed that uterine smooth muscle was with hyperplasia,muscular layer hypertrophy and disorder align of cells in model control group,and these pathological changes were improved in high and low dosage of Lichong decoction groups.Conclusion Lichongtang can significantly decrease serum sex hormone level such as E2,P,LH and improve the pathology of uterine tissue,it indicates that the anti-tumor effect of Lichongtang on uterine leiomyoma might be related,at least partly,to the adjustment of endocrine and enhancement of immune function.
