摘要
目的观察芪榔合剂对慢传输型便秘小鼠结肠Cajal间质细胞的影响,探讨其通便机制。方法用复方地芬诺酯制作实验性便秘小鼠模型,造模成功后随机分为便秘模型组、中药大剂量组、中药小剂量组,并设正常对照组,给予相应干预措施,共干预15天。观察小鼠排便时间、炭末推进率;光镜观察小鼠结肠Cajal细胞结构、免疫组化法观察小鼠结肠c-kit阳性表达及小鼠结肠c-kitmRNA阳性表达。结果与正常对照组比较,模型组首次排便时间明显延长,8 h、20 h排便总数明显减少,不排便动物数明显增多,小肠炭末推进率明显降低,结肠c-Kit阳性细胞数、c-kit mRNA表达率明显减少,差异均有统计学意义(P<0.05,P<0.01),说明造模成功;与模型组比较,中药大小剂量组首次排便时间明显缩短,8 h、20 h排便总数明显增多,小肠炭末推进率明显升高,结肠c-Kit阳性细胞数、c-kit mRNA表达率明显增加,差异均有统计学意义(P<0.05,P<0.01)。光镜下显示,与正常对照组比较,模型组Cajal间质细胞出现基膜溶解,数量减少,其与周围细胞之间的紧密连接也被破坏,细胞核有不同程度萎缩;与模型组比较,中药大小剂量组的Cajal间质细胞数量增加。结论芪榔合剂具有较好促进小鼠肠动力的作用,其通便机制可能为通过促进Cajal细胞的再生及修复而达到促进肠动力,从而实现通便的作用。
Objective To analyze the mechanism and observe the effects of "Qilang Deeoetion" on colon interstitial cells of Cajal in rats with slow transit constipation. Methods Compound Diphenoxylate was used to build the constipation model for 15 days. The rats were randomized into eonstipationtmodel group, Chinese medicine groups (high dose, small dose) and normal group. Each group corresponded to different intervening measures for 15 days. The defecation time, charcoal propulsion rate, the structure of colon interstitial Cajal eells, positive expressions of colon C-kit and C-kit mRNA were observed. Results Compared with normal group, the defecation time was prolonged significantly, the defecation was decreased at 8h and 20h, and the charcoal propulsion rate, C-kit positive expression and C-kit mRNA expression rate were significantly improved; compared with model group, the first defecation time in Chinese medicine groups was shorten, and the defecation was inereased at Rh and 20h, and the ehareoal propulsion rate, C-kit positive expression and C-kit mRNA expression rate were significantly improved, with significant differences( P 〈 0.05 ) ; compared with normal group, Cajal cells matrix decomposition and nueleus atrophy appeared, cells eonneetion was broken in model group ; compared with model group, the number of Cajal cells was increased. Conclusion "Qilang Deeoetion" can promote gastrointestinal motility, repair and regenerate the Cajal cells in rats to move the bowels.
