5-氟尿嘧啶对瘢痕疙瘩成纤维细胞MMPS及TIMPS表达的影响 被引量:4

摘要 目的研究5-氟尿嘧啶对体外培养的人类瘢痕疙瘩成纤维细胞MMP-1,MMP-2/TIMP-1,TIMP-2mRNA表达的影响,探讨5-氟尿嘧啶对瘢痕疙瘩的作用机制。方法组织块法培养人瘢痕疙瘩及正常皮肤组织成纤维细胞,用不同浓度的5-氟尿嘧啶作用于体外培养的人瘢痕疙瘩成纤维细胞,检测MTT值;利用半定量RT-PCR分析用药前后MMP-1,MMP-2,TIMP-1,TIMP-2 mRNA表达变化。结果 5-氟尿嘧啶在体外能明显降低人瘢痕疙瘩成纤维细胞TIMP-lmRNA的表达;对MMP-1,MMP-2,TIMP-2mRNA的表达无显著影响。结论 5-氟尿嘧啶可能通过降低TIMP-1的表达,从而使MMP-1/TIMP-1相对比例增加,达到促进胶原降解,减少瘢痕形成的作用。
