骨肉瘤组织中多药耐药基因mdr-1mRNA和p-糖蛋白的表达及其与临床病理特点关系的研究 被引量:11

The expression of the multidrug resistance gene mRNA and p-glycoprotein and its relative clinical and pathological research in human osteosarcoma

摘要 目的 研究多药耐药基因m dr - 1 m RNA 与p - 糖蛋白(p - gp) 在骨肉瘤的表达及与临床的关系。方法 40 例骨肉瘤及10 例正常骨组织冰冻切片,以地高辛标记m dr - 1 寡核苷酸探针及p - gp 单克隆抗体C219 分别进行原位杂交及免疫组化染色, 并对有关临床及病理指标综合分析。结果 骨肉瘤组m dr - 1 m RNA 和p - gp 的阳性表达率分别为85 % 和80 % ,正常骨组织相应值为10 % 和0 ,两组间比较差异有显著性意义( P< 0-05) 。此外,m dr - 1 m RNA 及p - gp 在骨肉瘤转移组、肿瘤较大、外科分期和病理分级较高的病例组中的表达水平高于无转移组、肿瘤较小、外科分期和病理分级较低的病例组( P< 0-05) 。而且,m dr - 1 m RNA 及p - gp 的表达在骨肉瘤标本中呈正相关( rs = 0-738 , P< 0-05) 。结论 人骨肉瘤恶性度高、对化疗不敏感、易转移与组织中存在m dr - 1 m RNA 及p - gp 有关, 两者表达水平与骨肉瘤的病理分级和外科分期及转移呈正比, 且两者表达呈正相关关系。原位杂交与免疫组化相结合是有效、简便地检测m dr - 1 Objective To study the expression of the multidrug resistance gene (mdr-1) mRNA and p-glycoprotein (p-gp) in human osteosarcoma and their relationship with clinical features Methods Digoxin labeling mdr-1 oligonucleotide probe and p-gp monoclonal antibody C219 were used to examine the expression of mdr-1 and p-gp in specimen frozen sections of 40 human osteosarcomas and 10 normal human bone tissue by in situ hybridization and immunohistochemical staining protocol Furthermore, their correlation with patients clinical manifestations and histopathological characteristics were analyzed Results The positive expression rates of mdr-1 mRNA and p-gp were 85%and 80%respectively in osteosarcoma, but only 10%and 0 in normal bone tissues;statistically the difference between osteosarcoma and normal bone tissues was significant (P< 0 05) Additionally the positive expression rates of mdr-1 mRNA and p-gp in patients with distant metastasis, large tumor, higher surgical stage and pathological grade increased obviously in comparion with those cases of non metastasis, small tumor and lower surgical stage and pathological grade (P< 0 05) Moreover, a significant positive correlation could be identified between mdr-1 mRNA and g-gp protein in osteosarcoma (rs=0 738, P< 0 05) Conclusion The study showed that the examination of mdr-1 mRNA and p-gp expression in osteosarcoma is of important significance in clinical treatment and research work The results also suggested that the combination of in situ hybridization with immunohistochemistry was an effective and simple method for the examination of the expression of mdr-1 gene
