摘要
硫酸二乙酯(DES)是一种能诱导生物体产生随机点突变的烷化剂。为了完善利用DES诱导家蚕突变的实验技术,采用不同剂量DES分别处理家蚕蛹、蛾、卵,检测DES对不同发育时期家蚕毒性的半致死剂量(LD50),结果表明DES对不同发育时期家蚕的LD50值存在显著差异:化蛹第5天的雌雄蛹分别为2 333.90μg/头和2 369.50μg/头;刚羽化雄蛾为156.39μg/头;蚕卵为13.23 mg/mL。研究结果可作为DES对家蚕的最适诱变剂量和家蚕诱变最佳发育时期选择的参考依据。
The alkylating agent diethyl sulfate(DES) can cause random point mutation in organisms.In order to improve the experimental techniques of using DES to induce mutation in silkworm(Bombyx mori),silkworm pupae,moths and eggs were treated with different doses of DES,and the medial lethal dose(LD50) of DES to silkworm at different developmental stages were calculated.The results showed that LD50 values of DES to silkworm at different developmental stages were significantly different.LD50 of DES to the 5th day female and male pupa was 2 333.90 and 2 369.50 μg per pupa respectively.That to the newly emerged moth and eggs was 156.39 μg per moth and 13.23 mg/mL respectively.These results provide good reference for determining the optimal dosage and the best developmental stage in using DES to induce mutation in Bombyx mori.
