摘要
目的观察丹红注射液对大鼠心肌缺血-再灌注(I-R)损伤中细胞凋亡的作用及其可能机制。方法 56只SD大鼠随机分为四组:假手术组(SH组,8只)、I-R组(16只)、丹红注射液组(DH组,16只)和丹红注射液+LY294002组(DH+LY组,16只)。图像分析系统计算梗死区面积,TUNEL法检测心肌细胞凋亡,Western blot法测定磷酸化Akt(p-Akt)表达。结果与I-R组相比,DH组梗死面积、细胞凋亡指数减少,p-Akt表达增加(P<0.01),而DH+LY组能逆转这些抗凋亡作用。结论丹红注射液减少大鼠I-R损伤中心肌细胞的凋亡,这可能与磷脂酰肌醇3激酶(PI3K)/Akt信号通路的活化有关。
Objective To investigate the effect and possible mechanism of danhong injection on the cell apoptosis in rats with myocardial ischemia-reperfusion(I-R) injury.Methods Fifty-six SD rats were randomly divided into four groups of SH(sham operated,8 rats),I-R(I-R model,16 rats),DH(injected with danhong 2 ml/kg before establishing I-R model,16 rats) and DH+LY(injected with danhong 2 ml/kg before establishing I-R model and treated with LY294002 0.3 ml/kg before reperfusion,16 rats).The infarction area was calculated by image analysis system and the myocardial cell apoptosis was detected by TUNEL method.The expression of phosphorylated-Akt(p-Akt) was determined by Western blot.Results Compared with group I-R,the infarction area and the cell apoptosis index were decreased,while the expression of p-Akt was increased in group DH(P〈0.01),which were all reversed in group DH+LY.Conclusion Danhong injection can reduce myocardial cell apoptosis in rats with I-R injury,which may be related with the activation of phosphatidylinositol 3-kinase(PI3K)/Akt signaling pathway.
