白藜芦醇对大鼠糖尿病性白内障抗氧化损伤的保护作用 被引量:10

摘要 目的探讨白藜芦醇对糖尿病性白内障大鼠抗氧化损伤的保护作用。方法 90只SD大鼠随机分为正常对照组、糖尿病模型组和白藜芦醇治疗组,每组各30只。模型组及白藜芦醇治疗组采用链脲佐菌素诱发糖尿病,白藜芦醇治疗组按400mg.kg-1体质量给予白藜芦醇灌胃,灌注后4周、8周、12周观察3组大鼠晶状体混浊度的变化情况;同时检测晶状体内超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathi-one peroxidase,GSH-PX)及过氧化氢酶(catalase,CAT)的变化;采用RT-PCR检测晶状体中铜锌超氧化物歧化酶(Cu-Zn superoxide dismutase,CuZnSOD)mRNA表达水平。结果正常对照组大鼠晶状体始终透明。灌注后12周白藜芦醇治疗组晶状体混浊度高于正常对照组(χ2=8.26,P<0.05),但与糖尿病模型组相比明显减轻(χ2=14.72,P<0.05)。糖尿病模型组大鼠晶状体中SOD、GSH-PX以及CAT活性分别为:(10.39±1.25)U·mg-1、(24.36±1.05)U·mg-1、(1.62±0.24)U·mg-1,明显低于正常对照组的(40.16±1.87)U·mg-1、(67.86±1.24)U·mg-1、(5.57±1.54)U·mg-1。与糖尿病模型组相比,白藜芦醇能增强SOD、GSH-PX和CAT活性,分别为(29.06±1.33)U·mg-1、(47.58±2.35)U·mg-1、(3.26±0.47)U·mg-1。此外,白藜芦醇还能上调糖尿病大鼠晶状体内CuZnSODmRNA的表达(P<0.05)。结论白藜芦醇能增加晶状体抗氧化酶活力,能从多个环节发挥对糖尿病白内障的抗氧化作用,从而延缓白内障的发生和发展。 Objective To investigate the oxidation resistance protection effect of resveratrol on diabetic cataract rats.Methods About 90 rats were randomly divided into normal control group,diabetic model and Resveratrol group,30 rats in each group.Diabetic cataract was induced by streptozotocin,and 400 mg·kg-1 of resveratrol was administrated by gavage.At the end of 4 weeks,8 weeks and 12 weeks after streptozotocin injection,eyeballs were isolated and cataract formation in all rats was recorded.The activity of superoxide dismutase(SOD),glutathion peroxidase(GSH-PX) and catalase(CAT) were detected by colorimetry.The mRNA level of Cu-Zn superoxide dismutase(CuZnSOD) was analyzed by RT-PCR.Results The crystalline lens were transparent in control group.After 12 weeks,the opacity in resveratrol group was higher than those in control group(χ2=8.26,P0.05),but was lower than those in diabetic model group(χ2=14.72,P0.05).The activities of SOD,GSH-PX and CAT in diabetic model group were(10.39±1.25)U·mg-1,(24.36±1.05)U·mg-1,(1.62±0.24)U·mg-1,obviously lower than(40.16±1.87)U·mg-1,(67.86±1.24)U·mg-1,(5.57±1.54)U·mg-1 in normal control group.Compared with control group,resveratrol could enhance activities of SOD,GSH-PX and CAT,which were(29.06±1.33)U·mg-1,(47.58±2.35)U·mg-1 and(3.26±0.47)U·mg-1.Resveratrol could also increase the mRNA expression level of CuZnSOD(P0.05).Conclusions Resveratrol could increase the antioxidase activity,and it plays an important role in many steps of antioxidation,thus inhibits or delays the generation and development of cataract.
