小鼠嗅鞘细胞培养的初步研究 被引量:2

摘要 目的:研究小鼠嗅鞘细胞(OECs)体外培养的不同方法,为获取数量多、活性好、形态优的嗅鞘细胞提供一定的实验基础。方法:4只出生2~3个月的BALB/c小鼠,经枕骨大孔取嗅球,剥离嗅球外周两层制成细胞悬液。采取未纯化培养对照组、一次差速贴壁结合阿糖胞苷(Ara-c)纯化与多聚左旋赖氨酸包被培养板培养组(纯化包被组)和一次差速贴壁结合Ara-c纯化与未用多聚左旋赖氨酸包被培养板培养组(纯化未包被组)分别培养小鼠OECs,观察其原代和传代培养的细胞生长状况及形态,并进行免疫细胞化学鉴定。结果:纯化未包被组OECs早中期无论数量还是形态都优于纯化包被组,而未纯化对照组的OECs早中期数量明显优于纯化组。结论:一次差速贴壁结合Ara-c纯化未包被进行小鼠OECs培养是一种简捷有效地方法。
