摘要
蛋白质翻译后修饰几乎参与了细胞所有的正常生命活动,并发挥十分重要的调控作用,目前已成为国际上蛋白质研究的一个极其重要的领域。常见的蛋白质翻译后修饰包括类泛素化、乙酰化、磷酸化、甲基化、糖基化等。傅里叶变换-离子回旋共振质谱(FT-ICR MS)具有超高分辨率、高质量测量准确度、质量范围宽、速度快、性能可靠等显著优点,在确定蛋白质翻译后修饰位置以及修饰的结构研究方面具有独特优势,对其在蛋白质翻译后修饰方面的应用进行了较全面的综述和讨论。
Protein post-translational modification involved in nearly all cells in the normal life activities,and play a very important role in the regulation,which has become an extremely important area for studying of the protein.Common types of protein post-translational modifications include ubiquitination,acetylation,phosphorylation,methylation,glycosylation and so on.Fourier transform-ion cyclotron resonance mass spectrometry(FT-ICR MS) has shown powerful performance due to the merits of ultrahigh resolution,high accuracy,broadband,fast,reliable performance.FT-ICR MS has unique advantages in determining the location and modified structure of protein post-translational modification.This work reviewed and discussed the application of FT-ICR MS in protein post-translational modification.
