摘要
目的探讨辛伐他汀对大鼠随意皮瓣成活的影响及其可能的作用机制。方法健康清洁级雄性SD大鼠56只,6周龄,体重(220±20)g,随机分为实验组及对照组各28只。背部制作随意皮瓣模型后,实验组大鼠通过腹腔注射辛伐他汀5mg/kg,对照组给予等容积生理盐水腹腔注射,每天1次。7d后,大体观察两组皮瓣的成活情况并计算成活率,光镜下观察两组皮瓣形态及病理变化,免疫组化法检测近、中、远三段微血管密度(MVD)。结果术后7d,实验组皮瓣成活率及皮瓣中段MVD均较对照组高(P<0.05),实验组皮瓣血管再生明显多于对照组。结论辛伐他汀可提高随意皮瓣成活率,其机制可能与促进血管新生有关。
Objective To investigate the effects of simvastatin on the survival of the random skin flaps in rats and its possible mechanisms.Methods 56 SD rats of both males aged 6 weeks weighing 220±20g were randomly divided into two groups with 28 rats in each group,and a random dorsal skin flap was established in each animal.In the experimental group each rat was given a intraperitoneal injection with simvastatin(5mg/kg) and the control group animals each received the same volume of saline also by intraperitoneal injection,Once a day for 7 days.The viable situation and survival rate of all flaps in the two groups were observed,meanwhile pathological changes were observed with light microscope,and immunohistochemical test was performed to detect the microvessel density(MVD).Results At 7 days after operation,the flap survival rate in the experimental group was remarkably higher than that in the control group.The MVD in the middle segment of the flap was greater in the experimental group than that in the control group(P 0.05).Histological semiquantitative analysis showed angiogenesis in flaps treated with simvastatin was much greater than that in the control group.Conclusion Simvastatin can increase the survival rate of random skin flaps in rats,which may be related to the promotion of angiogenesis.
