枸杞多糖对双酚A暴露小鼠生精细胞Caspase-3、Bcl-2和Bax蛋白表达的影响 被引量:13

摘要 研究枸杞多糖(LBP)对双酚A(BPA)暴露小鼠睾丸生精细胞中Caspase-3、Bcl-2和Bax凋亡蛋白表达的影响。将50只成年雄性昆明小鼠随机分为A、B、C、D、E共5组,每组10只。除正常对照组(A组)注射等量橄榄油外,其余4组小鼠分别腹腔注射20mg.kg-1的BPA,连续7d,建立生精损伤模型。同时C、D、E组小鼠分别灌服7d不同剂量的LBP(50、100、200mg.kg-1),正常对照组(A组)和模型组(B组)小鼠灌服等量生理盐水。制备组织切片观察睾丸组织病理学变化,免疫组化法测定睾丸组织Caspase-3、Bax和Bcl-2凋亡蛋白的表达。结果显示,BPA可极显著增加睾丸生精细胞Caspase-3和Bax的阳性细胞数量(P<0.01),降低Bcl-2的表达(P<0.05)。补充不同剂量LBP后,Caspase-3的阳性表达均极显著低于模型组(P<0.01)。200mg.kg-1 LBP组生精细胞Bax的阳性细胞数量极显著低于模型组(P<0.01);Bcl-2的表达随LBP剂量的增加而提高,其中200mg.kg-1 LBP组阳性表达极显著高于模型组(P<0.01),Bcl-2/Bax比值也随着LBP剂量的增加而上升。结果表明,枸杞多糖通过调节凋亡相关基因的表达,抑制生精细胞凋亡,从而缓解双酚A引起的雄性生殖损伤。 The aim of this experiment was to study the effects of LBP on the expression of Caspase-3, Bel-2 and Bax proteins in spermatogenie cells of mice. Fifty adult male Kunming mice were randomly divided into five groups, which were A, B, C, D and E, with 10 miee in each group. Ex- cept the mice in group A which were injected with olive oil as control group, the mice in other four groups were intraperitonealy injected with 20 mg.kg-1 BPA for 7 days. Meanwhile, the mice in groups C, D, E were given different doses of LBP (50, 100 and 200 mg.kg-1) by intragastrie administration consecutively for one week, the mice in control (A) group and model (B) group were treated with physiological saline. Histopathologieal changes of the testicular tissue were ob- served, immunohistoehemieal technique was applied to detect the expression of Caspase-3, Bcl-2 and Bax proteins. Results showed that BPA can enhance the expression of Caspase-3 and Bax proteins (P〈0.01), diminish the expression of Bcl-2 (P〈0.05). After supplement with differ- ent doses of LBP, the expression of Caspase-3 were all significantly deereased in C, D and E groups than that in the model group (P〈0.01). With the dose of LBP increasing, the expressionof Bax was significantly decreased and the expression of Bcl-2 gradually increased than that in the model group especially in the 200 mg .kg-1 LBP group (P〈0.01). Compared with the model group, Bcl-2/Bax ratio also increased obviously in groups B, C and D. Results suggest that LBP can restrain spermatogenic cell apoptosis through regulating the expression of apoptosis-related genes, thus alleviate the reproductive damage induced by BPA on adult male mice.
