摘要
目的:研究黄芪注射液对格列美脲在大鼠体内药动学的影响。方法:将大鼠随机分为对照组和联合用药组,对照组单独灌胃100mg·kg-1的格列美脲混悬液,联合用药组静脉注射8mL·kg-1黄芪注射液5min后灌胃100mg·kg-1的格列美脲混悬液。不同时间点于股动脉采集血样,经高效液相色谱法测定格列美脲的血药浓度,用DAS 2.0软件计算药动学参数后进行统计学分析。结果:对照组t1/2β、AUC分别为(44.58±2.97)h、(10.91±2.67)mg·h·L-1,联合用药组t1/2β、AUC分别为(65.67±5.37)h、(15.14±4.35)mg·h·L-1。结论:黄芪注射液对格列美脲的药动学有显著性影响。
OBJECTIVE: To research the effects of Astragalus membranaceus injection on the pharmacokinetics of glimepiride in rats. METHODS: Rats were randomly divided into control group and drug combination group. Control group was given glimepiride suspension 100 mg·kg^-1intragastrically. Drug combination group was given glimepiride 100 mg·kg^-1 intragastrically 5 min after intravenous injection of A, membranaceus injection 8 mL·kg^-1. Blood sample of femoral artery were collected at different time points. The blood concentration of glimepiride was determined by HPLC. Pharmacokinetie parameters was calculate using DAS 2.0 software and analyzed statistically. RESULTS: The t1/2β and AUC were (44.58 ± 2.97) h and (10.91 ± 2.67) mg·h·L^-1 in control group, and those were (65.67 ± 5.37) h and (15.14 ±4.35) mg·h·L^-1 in drug combination group. CONCLUSION: A. membranaceus injection has significant effects on the pharmacokinetics of glimepiride in rats.
