摘要 目的探讨Twist和HIF-1αmRNA在前列腺癌组织中表达及临床意义。方法应用原位杂交技术分别检测30例前列腺增生组织、30例前列腺上皮内瘤变组织及70例前列腺癌组织Twist、HIF-1αmRNA表达情况。结果①Twist、HIF-1αmRNA在30例前列腺增生组织、30例前列腺上皮内瘤变组织及70例前列腺癌组织中的表达分别为13.3%、46.6%、65.7%和16.7%、50.0%、71.4%,差异有统计学意义(χ2=23.15,P<0.01;χ2=25.57,P<0.01);Twist、HIF-1αmRNA在同组间表达差异无统计学意义(P>0.05)。②Twist及HIF-1αmRNA的表达均与前列腺癌的Gleason分级、临床分期、骨转移及组织坏死相关(P<0.05),而与年龄无关(P>0.05)。③Twist、HIF-1αmRNA在前列腺癌组织中过度表达,且两者之间呈正相关(r=0.34,P<0.01)。结论 Twist、HIF-1αmRNA在前列腺癌发生发展过程中起着重要作用,两者表达检测有利于更好地评估前列腺癌的生物学行为,提高对患者预后判断的准确性。 Objective To investigate the expressions of twist and HIF-1α mRNA in adenocarcinoma of prostate and their sig- nificance. Methods The expression of twist and HIF-1α mRNA were detected on prostatic hyperplasia( 30 cases), prostatic in- traepithelial neoplasm(30 cases) and adenocarcinoma of prostate(70 cases) by in situ hybridization. Results (2)The positive rates of twist and HIF-1α mRNA in prostatic hyperplasia,prostatic intraepithelial neoplasm and adenocareinoma of prostate were 13.3% ,46.6% ,65.7% and 16.7% ,50.0% ,71.4% ,respectively,there was statistical difference(x2 = 23.15 ,P 〈0.01 ;X: = 25.57 ,P 〈0.01 ) ;There was no statistical difference between twist and HIF-1α mRNA in same group. (2)The expression of twist and HIF-1α mRNA was related to Gleason grade, clinical stage, bony metastasis and tissues necrosis (P 〈 0.05 ), but not to age ( P 〉 0.05 ). (3)The over-expressions of twist and HIF-1α mRNA in adenocarcinoma of prostate showed positive relative relation- ship between them( r = 0.34 ,P 〈 0.01 ). Conclusion Twist and HIF-1α mRNA played an important role in the occurrence and development of adenoearcinoma of prostate, and it would provide valuable information to evaluate biological behaviour of adenocarcinoma of prostate and improve the patient ,s prognosis accuracy.
