Expression and significance of NF-κB and MIF in endometriosis

摘要 目的:探讨NF-κB、MIF与子宫内膜异位症(EMs)发病的关系,及在子宫内膜异位症表达的相关性。方法:采用免疫组化(SABC法)分别检测NF-κB、MIF在35例EMs在位内膜、60例EMs异位内膜及30例正常子宫内膜(对照组)中的表达;ELISA法检测TNF-α、PDTC+TNF-α干预后子宫内膜基质细胞(ESC)上清液中NF-κB、MIF的浓度,并统计分析检测结果。结果:(1)NF-κB、MIF在3组内膜中均有表达,以异位内膜的表达最强;(2)NF-κB、MIF的表达与月经周期无关,其在3组内膜的表达有相关性,相关系数为0.875、0.882、0.927;(3)NF-κB、MIF在经TNF-α、PDTC+TNF-α处理过的ESC上清液中有表达,TNF-α促进二者表达,而PDTC可抑制此作用,且两者表达有相关性,相关系数为0.947。结论:NF-κB、MIF在EMs内膜高表达,MIF可能通过NF-κB途径在EMs发病机制中起作用。 Objective:To evaluate the relationships between NF-κB and MIF with en- dometriosis(EMs) and the expression correlation between NF-κB and MIF in EMs. Methods: NF-KB and MIF protein expression were detected by Immunohistochemistry(SABC) in 35 cases of eutopic endometrium and 60 cases of ectopic endometrium in endometriosis and 30 cases of normal endometrium as control. The expressions of NF-κB and MIF were detected by enzyme- linked immunosorbent assay (ELISA)in the culture supernatant which responsed to TNF-α, PDTC+ TNF-α. The results were analyzed statistically. Result:( 1 )The expressions of NF-κB and MIF were found in the endometrium of all three groups. It was expressed most in eetopic en- dometriotic tissue. ( 2 ) The expressions of NF-κB arid MIF in endometriosis have no cyclic change. There are "correlations" between NF-KB and MIF and the correlation coefficient are O. 875,0. 882 and 0. 927. (3) NF-κB and MIF were detected in the culture supernatant of the ESC which responsed to TNF-α and PDTC+ TNF-α. TNF-α can promote the effect, PDTC can inhibit the effect. There are positive correlation between NF-KB and MIF and the correlation co- efficient was 0.947. Conclusion:The expression of NF-κB and MIF is high in endometrium of EMs patients. MIF may play an important role via NF-κB pathway in human endometriosis.
