摘要
目的建立RP-HPLC法测定虎杖提取物中白藜芦醇苷、白藜芦醇、大黄素和大黄素甲醚的含量。方法采用Kromasil C8色谱柱,以甲醇-水为流动相进行梯度洗脱。流速为1 mL/min;检测波长为290nm;进样量为20μL;柱温为30℃。结果虎杖提取物中白藜芦醇苷、白藜芦醇、大黄素和大黄素甲醚分别在10.54~52.7μg/mL,4.5~22.5μg/mL,4.92~49.2μg/mL和3.51~35.1μg/mL范围内与相应峰面积呈良好线性关系,平均加样回收率分别为97.70%、97.20%、102.6%、100.0%(n=6),RSD值分别为2.36%、1.89%、1.23%、3.67%。结论该方法快速、准确、简便,可用于虎杖提取物的质量控制。
Objective To develop a simple and rapid RP-HPLC method for simultaneously determination of polydatin,resveratrol,emodin and physcion in extract from Polygonum cuspidatum.Methods The HPLC method was performed on a Kromasil C8 at 30 ℃,and the mobile phase was methanol-water with gradient elution at a flow rate of 1 mL/min.The detection wave length was 290 nm,and 20 μL was injected.Results The linear range of polydatin,resveratrol,emodin and physcion were 10.54-52.7 μg/mL,4.5-22.5 μg/mL,4.92-49.2μg/mL and 3.51-35.1 μg/mL,respectively.The average recovery ratio were 97.70%,97.20%,102.6% and 100.0%,with the relative standard deviations(RSD)of 2.36%,1.89%,1.23% and 3.67%,respectively.Conclusion The method proved to be rapid,accurate and simple,suitable for the quality control of extract of Polygonum cuspidatum.
