摘要
目的探讨Toll样受体2(TLR 2)和Toll样受体4(TLR 4)在人颈动脉硬化斑块中的表达及其与斑块形成的关系。方法根据术后组织病理学报告选择性收集36例新鲜颈动脉硬化斑块,其中软斑组12例,硬斑组12例,混合斑组12例,以及12例正常脾动脉组织作为对照组,采用R T-PCR检测标本组织中TLR 2及TLR 4 mR NA的表达水平;应用免疫组化检测TLR 2和TLR 4蛋白在标本组织中的表达。比较不同性质颈动脉硬化斑块组织TLR 2和TLR 4的mR NA及蛋白表达水平。结果 R T-PCR结果显示颈动脉硬化斑块组织中TLR2和TLR4相对水平分别为(0.926±0.047)和(1.040±0.056),显著高于脾动脉组织的(0.646±0.033)和(0.691±0.031)(P<0.05);免疫组化结果显示TLR2和TLR4在颈动脉硬化斑块中相对水平分别为(135.295±17.787)和(130.368±14.969),显著高于脾动脉组织的(82.396±6.668)和(98.898±9.089)(P<0.05);且TLR 2和TLR 4mR NA及蛋白表达上调水平相关,TLR 2和TLR 4mR NA及蛋白表达水平与斑块性质有关,差异有统计学意义(P<0.05)。结论 TLR2及TLR4在颈动脉硬化斑块组织中高表达;TLRs信号通路可能促进了颈动脉硬化程度的进展。
[ Objective ] To investigate the expressions of toll-like receptor 2 and toll-like receptor 4 in human carotid atherosclerotic plaques. [Methods] 36 cases of human fresh carotid atherosclerotic plaque specimens and 12 specimens of patients with splenic artery (control group) were selective collected after operation, according to the postoperative histopathologic report, among them soft spot group in 12 cases, hard spot group in 12 cases, mixed spot group 12 cases, the RT-PCR detection specimens in the organization TLR2 and TLR4 mRNA expression level; application immunohistochemical detection TLR4 and TLR2 in the organization is the expression of protein samples. compare different nature of the carotid atherosclerotic plaques organization TLR2 and the TLR4 mRNA and protein expression level. [ Results ] The results show that carotid atherosclerotic plaques organization TLR2 and TLR4 relative level is 0.926±0.047 and 1.040±0.056, significantly higher than normal splenic artery organization of (0.646± 0.033) and (0.691±0.031)(P 〈0.05); immunohistochemical results show that TLR2 and TLR4 carotid atheroselerotie plaques in the relative level were (135.295±17.787) and (130.368±14.969), significantly higher than normal splenic artery organization of (82.396~6.668) and (98.898±9.089)(P 〈0.05); TLR2 and TLR4mRNA and raise the level of protein expression related, TLR2 and TLR4 mRNA and protein expression level and plaques on nature, differences have statistical significance (P 〈0.05). [ Conclusion ] TLR2 and TLR4 carotid atherosclerotic plaques organization in high expression; TLRs signaling pathways may promote the carotid artery sclerosis degree of progress.
