呼吸道合胞病毒毛细支气管炎患儿外周血CD4^+CD25^+调节性T细胞及IL-10、TGF-β检测及意义 被引量:6

摘要 目的观察呼吸道合胞病毒(respiratory syncytial virus,RSV)毛细支气管炎(毛支)患儿外周血CD4+CD25+调节性T细胞(Treg)及其分泌细胞因子IL-10、TGF?β的水平,探讨Treg与RSV毛支发病的关系。方法采集31例RSV阳性毛支患儿外周血,采用流式细胞仪检测外周血Treg细胞百分率,ELISA法检测血浆IL-10、TGF?β的水平。27例非RSV感染的普通肺炎患儿作为阳性对照,24例健康儿童为正常对照组。结果 RSV毛支患儿外周血Treg百分率(10.71±1.11)%显著低于肺炎组(12.73±1.35)%及正常对照组(13.24±1.13)%(P<0.01);RSV毛支组血浆IL-10水平(89.83±9.43)ng/L显著低于肺炎组(116.40±14.10)ng/L与正常对照组(124.22±18.38)ng/L(P<0.01);RSV毛支组TGF?β水平(1 019.17±353.32)ng/L显著低于肺炎组(1 979.50±462.30)ng/L与正常对照组(2 139.88±484.75)ng/L(P<0.01)。结论 RSV毛支患儿外周血存在Treg细胞数量及功能的不足,可能是引起RSV毛支发作的重要因素之一。 Objective To observe the level of CD4+CD25+regulatory T cells(Treg)and cytokines IL-10 and TGF-β in peripheral blood in infants with bronchiolitis caused by respiratory virus(RSV),and discuss the relationship between Treg and RSV bronchiolitis.Methods The percentage of Treg in peripheral blood in 31 infants with RSV bronchiolitis,27 infants with non-RSV pneumonia and 24 healthy infants were detected by flow cytometry.The level of their cytokins IL-10 and TGF-β in peripheral blood was measured by ELISA.Results The percentage of Treg in peripheral blood in infants with RSV bronchiolitis(10.71±1.11)% was significantly lower than these in infants with non-RSV pneumonia(12.73±1.35)% and healthy infants(13.24±1.13)%(P0.01).The level of IL-10 and TGF-β in peripheral blood in infants with RSV bronchiolitis(89.83±9.43,1 019.17±353.32)ng/L were significantly lower than these in infants with non-RSV pneumonia(116.40±14.10,1 979.50±462.30)ng/L and healthy infants(124.22±18.38,2 139.88±484.75)ng/L(P0.01).Conclusion There are a decrease of the percentage of Treg and their insufficient function in peripheral blood in infants with RSV bronchiolitis,Treg may be an important factor of the pathogenesis of RSV bronchiolitis.
