摘要
目的探讨骨髓单个核细胞移植对创伤性脑损伤(TBI)大鼠脑源性神经营养因子(BDNF)表达的影响。方法 SD大鼠随机分为假手术组、脑外伤组(TBI组)和TBI+单核细胞移植治疗组。建立自由落体TBI大鼠脑损伤模型,取骨髓分离纯化单个核细胞(5×106/mL),经颈动脉注射移植(单核细胞移植治疗组大鼠每只注射1mL)。各组动物分别于TBI后第1、3、7、10d进行神经功能缺损评分(NSS)。TBI后7d取损伤处脑组织行RT-PCR检测BDNF mRNA表达,TBI后10d取损伤处脑组织行原位杂交和免疫组化染色,确定BDNF mRNA和蛋白的细胞定位,计数BDNF阳性细胞数量变化。结果骨髓单个核细胞移植治疗明显减少TBI大鼠NSS评分,与TBI组比较,差异有统计学意义(P<0.05)。BDNF主要分布于皮质神经元,移植组BDNF mRNA和蛋白表达水平均明显上调(P均<0.05)。结论单个核细胞移植能改善TBI大鼠神经功能,减少神经功能缺损。其机制可能与BDNF表达上调有关。
Objective To investigate the effect of single neucleated cells transplantation on the neurological function and brain derived neurotrophic factor(BDNF) expression in traumatic brain injury(TBI) rats.Methods TBI rats were established by hammer fall method from 30 cm height with 50 g weight.Single neucleated cells/karyocytes from bone marrow were separated and prepared by ficoll medium,then transplanted into pericontusional tissues in the brain.Neurological function severity scores(NSS) were recorded at 1,3,and 7 days post operation(dpo).The pericontusional tissues were harvested at 7 dpo to analyze the BDNF localization,expressional level.Results Single neucleated cell transplantation decreased NSS significantly,compared with TBI rats without cell implantation(P0.05).BDNF expression was upregulated and mainly found in neurons.Conclusion The present study showed the single karyocytes transplantation could improve neurological function and the mechanism is possibly linked to the BDNF expression.
