慢性氟中毒大鼠神经细胞凋亡的研究 被引量:28

摘要 目的研究慢性氟中毒对神经细胞的作用机制,特别是氛中毒与细胞凋亡的关系。方法采用末端标记法和流式细胞仪观察法,测定了慢性氟中毒鼠脑组织中神经细胞的凋亡情况。结果TUNEL检测显示,在大鼠慢性氟中毒模型中,可见到明显的DAB着染的阳性凋亡细胞,而且这种表达具有选择性,仅出现在海马CA4区。流式细胞仪显示,正常脑组织中的神经细胞即可测得凋亡峰;慢性氟中毒组脑组织中的凋亡峰明显高于正常对照组,但不同部位之间没有显著差异。结论在慢性氟中毒鼠的脑组织中有神经细胞发生凋亡的倾向,与病理改变结果十分相似。可能在慢性氟中毒过程中不只存在一种神经细胞损伤形式,而是退行性改变与凋亡并存。 Objective Study the action mechanism of chronic fluorosis in neurones. Methods Terminal deoxyribo-nucleotide transferase-mediated dUTP-biotin nick end labeling (TUNEL ) and flow cytometry (FCM) were used to observe changes of apoptosis cells of cerebral in chronic fluorosis in rats. Results TUNEL results show:There are apoptosis cells of DAB positive stain and the expression is not at random. It is only appear in hippocampus CA4 region;FCM results show: The percentage of DNA fragmentation are increased distinctly in the cerebral of chronic fluorosis in rats,but there are not discrepancy notability among different cerebral regions. Conclusions There is a tendency of apoptosis in neurone of chronic fluorosis in rats. It is most similitude with change results of pathology. It is not likely that only one form of neruone damage exist in the process of chronic fluorosis. There are recessive change and apoptosis in the process at the same time.
