摘要 针对大鲵(Andrias davidianus)性腺发育的不同步现象,为提高受精率,对其精液特性和保存条件进行初步研究。人工辅助采集精液,对精液量、酸碱度、密度、活力等基本特性进行常规测定,分别对保护液、光照、温度等保存条件进行了分组研究试验。结果表明,亲鲵的精液量最多可达12mL/kg,pH值6.7~7.0,呈微酸状态;精子密度215万~1075万/mL,精子活力为0.7~0.9。离体保存条件下,室温暗环境(20lx)较光环境(400lx)可延长存活时间0.5h,暗环境低温(0~4℃)较室温(21~22℃)可延长存活时间3d,低温暗环境下原精液中添加保护液比对照组精子存活时间延长6~12h。 Gonadal development of Andrias davidanus not synchronized, semen characteristics and preservation conditions of A. davidanus were studied targeting the increase of fertility rate. Semina were collected by artificial assistance approach, and a regular assay was conducted on the basic features such as semen volume, pH, density, vitality, etc. Preservation conditions, namely, protection medium, illumination and temperature, were also stud- ied. The results showed that semen volume of pro-male A. davidanus was up to 12 mL/kg, pH 6.7 -7.0 of subac- id condition; and that the semen density was(2.15 -10.75 )million/mL, sperm motility 0.7 -0.9. In vitro con- servation conditions, dark environment at ambient illuminance (20 Ix). The result also indicated that the survival time of the semina was 0.5 h longer than those conserved in the luminous environment (400 Ix) ; low temperature (0 -4℃ ) could extend the survival time of the semina 3d longer than those at room temperature (21 -22℃) in dark environment; in the cold dark environment, semina with protection medium added in could extend the survival time 6 - 12 h longer than the control group.
