摘要
目的观察养肝益水颗粒对12周龄自发性高血压大鼠(SHR)血浆血管紧张素Ⅱ(AngⅡ)、转化生长因子β1(TGF-β1)、结缔组织生长因子(CTGF)含量和肾皮质AngⅡ、TGF-β1、CTGF mRNA表达的影响,以进一步探讨养肝益水颗粒发挥肾脏保护的可能作用机制。方法将40只12周龄雄性SHR大鼠随机分为模型组、贝拉普利组[1.8mg/(kg·d)]、养肝益水颗粒低剂量组[5.4g/(kg·d),简称低剂量组]、养肝益水颗粒高剂量组[27g/(kg·d),简称高剂量组],每组10只;并设Wistar-Kyoto大鼠(WKY)10只作为正常对照组;模型组和正常对照组给予等量生理盐水灌胃,每天1次,共6周。检测各组大鼠血浆AngⅡ、TGF-β1、CTGF浓度和肾皮质AngⅡ、TGF-β1、CTGF mRNA的表达。结果模型组,低、高剂量组血浆AngⅡ、TGF-β1、CTGF含量显著高于正常对照组(P<0.01),贝拉普利组与正常对照组比较,差异无统计学意义(P>0.05);贝拉普利组与高剂量组血浆AngⅡ、TGF-β1、CTGF含量显著低于模型组(P<0.01,P<0.05),低剂量组与模型组比较,差异无统计学意义(P>0.05);贝拉普利组及高、低剂量组肾皮质AngⅡ、TGF-β1、CTGFmRNA的表达显著低于模型组(P<0.01,P<0.05);高剂量组优于贝拉普利组和低剂量组(P<0.01,P<0.05)。结论养肝益水颗粒主要通过降低肾脏局部AngⅡ、TGF-β1及CTGF的表达而发挥肾脏保护作用。
Objective To study the effect of Yanggan Yishui Granule(YGYSG) on 12-week-old spontaneously hypertensive rats(SHR) on plasma angiotensin Ⅱ(AngⅡ) ,transforming growth factor β1(TGF-β1) ,connective tissue growth factor(CTGF) content and renal cortical AngⅡ,TGF-β1,CTGF mRNA expressions,and to further explore the mechanism of YGYSG in the kidney protection.Methods Forty 12-week-old male SHR were randomized into the model group,the Bella Plymouth group(at the daily dose of 1.8 mg/kg) ,the YGYSG low dose group(at the daily dose of 5.4 g/kg) ,and the YGYSG high dose group(at the daily dose of 27 g/kg) ,10 in each group.Another ten Wistar-Kyoto(WKY) rats were included as the control group.Equal volume of normal saline was given to SHR by gastrogavage in the model group and the normal control group,once a day for six weeks.The plasma AngⅡ,TGF-β1 and CTGF concentrations,and the renal cortical AngⅡ,TGF-β1,CTGF mRNA expressions were measured in each group.Results Bella Plymouth was superior to YGYSG in reducing plasma AngⅡ,TGF-β1 and CTGF,while the high and low dose YGYSG showed no significant difference(P0.05) .In reducing plasma AngⅡ,TGF-β1 and CTGF mRNA expressions,low doses YGYSG was similar to the role of Bella Plymouth,and the role of high dose YGYSG was superior to low dose YGYSG and Bella Plymouth.Conclusion YGYSG played a role in kidney protection mainly through reducing the AngⅡ,TGF-β1 and CTGF expressions of kidney.
