摘要
研究了AB-8大孔树脂对豆腐柴根提取物中总黄酮静态和动态的吸附性能。利用分光光度法测定样品中黄酮含量,考察提取液浓度、pH值、乙醇洗脱体积分数、上样液流速等实验条件对吸附结果的影响。结果表明,AB-8大孔树脂对豆腐柴根中总黄酮具有较好的吸附性能。静态吸附最佳条件:上样液浓度为0.0275mg/mL、pH 7、70%乙醇洗脱,总回收率为65.45%;动态吸附的最佳条件:浓度为0.0275mg/mL、pH为5、流速为1mL/min,90%的乙醇洗脱条件下,总回收率为47.27%。
The static and dynamic adsorption performances of macorporous resin AB-8 on total flavonoids from Premna microphylla Turcz root extraction were investigated.The content of total flavonoids in samples was determined by spectrophotometry.The influences of the sample concentration,value of pH,volume fraction of ethanol elution and flow rate on adsorption results were discussed.The results showed that AB-8 resin had good adsorption capacity on flavonoids from Premna microphylla Turcz root extraction,and the optimum static adsorption conditions were as follows:the sample concentration was 0.0275 mg/mL at pH 7 with 70% ethanol aqueous as elution,and under such conditions,the recovery of flavonoids was 65.45%,while the recovery of total flavonoids of 47.27% was obtained by the following optimum dynamic adsorption conditions as follows:the sample concentration of 0.0275mg/mL,pH of 5,flow rate of 1mL/min and ethanol aqueous of 90% as elution.
