摘要
目的研究诱导型一氧化氮合酶(iNOS)在先天性肾积水肾组织的表达和分布情况,探讨它在先天性肾积水发病机制中的作用。方法采用免疫组织化学技术SABC法对36例先天性肾积水患者(病例组)、20例肾肿瘤患者正常的肾组织(对照组)进行iNOS染色,运用彩色图像分析系统测定其表达的积分光密度并作定量分析比较,同时与肾纤维化程度进行相关性分析。结果 iNOS主要表达于肾小管上皮细胞胞质、肾小球系膜细胞。积水肾组与对照组相比,iNOS表达的积分光密度明显下降,差异有统计学意义,P<0.01。积水肾组iNOS的表达与肾纤维化程度呈负相关,r=-0.649,P<0.001。结论 iNOS在积水肾表达减少加重了肾间质纤维化,iNOS所产生的一氧化氮在其发病过程中起保护作用。
[Objcetlve] To study the role of inducible nitric oxide synthase (iNOS) in the pathogenetic mecha- nism of congenital hydronephrosis. [Methods ] Immunohistochemical staining (SABC method) was performed to de- tect the iNOS expression in 36 kidney tissues of congenital hydronephrosis and 20 normal control kidney tissues ob- tained from renal neoplasm. The quantitative analysis was made by image analysis technique. Correlations between the iNOS expression and the degree of renal fibrosis were investigated. [ Results ] Immunoreactivity to iNOS was predominantly seen in epithelial cells of nephric tubule and mesangial cells. The iNOS expression was significantly decreased in congenital hydronephrosis group compared to normal group, P 〈0.01. A negative correlation was found between the iNOS expression and the degree of renal fibrosis (1" =-0.649, P 〈0.001). [ Conclusions] The decreased expression of iNOS aggravates the degree of renal fibrosis. NO produced by iNOS may play a protective role in the pathogenesis of congenital hydronephrosis.
