奶牛乳腺蛋白组分析中蛋白质分步提取方法的建立

摘要 全息蛋白质样品提取是蛋白组研究的先决条件,采用3种溶解性质不同的裂解液分步提取奶牛乳腺蛋白组,分别进行双向聚丙烯酰胺凝胶电泳分离,PDQUest 7.4分析凝胶图谱,发现其与常规方法相比,具有蛋白质提取率高、双向电泳(2-DE)分辨率高等优点。 The total sample preparation is a prerequisite for proteomic analysis.A three-step sequential protein extraction method was applied in proteomics analysis:using three kinds of solutions with differential solubility to replace one kind of solution.Proteins were separated by IEF and SDS-PAGE.And the patterns were analyzed with PDQUest 7.4 software.The method improved the efficiency of extraction and separation of the proteins compared with method of one solution.
