摘要
目的 观察雷公藤多甙对小肠移植急性排斥反应的治疗作用。方法 选用近交系大鼠F344/N 和Wistar/A 进行全小肠异位移植,实验分3 组,第1 组:非治疗对照组(F344Wistar);第2组:CsA 治疗对照组[F344Wistar+ Csa(6mg,肌肉注射)] ;第3 组:雷公藤多甙治疗组[F344Wistar+雷公藤多甙(50 mg/kg,灌胃)]。观察动物存活及一般状况,并于术后第3 、5 、7d 取移植肠标本进行病理学检查。结果 第1 组大鼠存活时间(6.83 ±0 .41) d,第3 组大鼠存活时间为(8.00 ±0 .63) d,二者间有极显著差异(P< 0 .01),第2 组大鼠存活均超过10 d,并于10 d 时处死。病理检查显示第1 组术后第3 d 发生轻度排斥,第5d 发生中度排斥,第7d 发生重度排斥;第2 组除部分标本在术后第5 、7d 发生轻度排斥外,无排斥征象;第3 组与第1 组改变相似,但排斥的病理改变发生较晚、较轻。结论 对小肠移植急性排斥的治疗,在50 mg/kg 剂量水平上单独使用雷公藤多甙,其免疫抑制作用是明确的,但疗效有限。
Objective The aim of the study is to assess the efficacy of Tripterygium wilfordii (TW) on the acute rejection in small bowel transplant.Method Heterotopic small bowel transplants were carried out using inbred F344 (donor) and Wistar (recipient) rats. The recipients were divided into three groups:1) allograft control group, 2) cyclosporin A treated group (6mg/kg, im) and 3) TW treated group (50mg/kg, enteral). Post transplant clinical course and survival rates were recorded. The grafts were harvested on post operative day 3, 5 and 7. The specimens were subjected to histological examination.Results The survival duration of group 3 ( 8.00 +/- 0.63 days) was significantly longer than that of the group 1 ( 6.83 +/- 0.41 days) ( P < 0.001 ). The average survival duration of group 2 rats was more than 10 days. The histology examination of the transplanted bowel specimen from group 1 rats showed mild, moderate and severe rejection on post operative day 3, 5 and 7. Some of the specimens from group 2 showed mild rejection on day 5 and 7. Group 3 specimens showed similar changes as group 1 except that the changes were milder and occurred later.Conclusion Tripterygium wilfordii, administered at the dosage of 50mg/kg, has limited but definite immuno suppression effect on acute rejection in small bowel transplantation in rats.
