摘要
目的应用基于ITO叉指电极微流芯片的电化学测菌法,探讨该方法检测变形链球菌的可行性。方法利用基于ITO叉指电极微流芯片的电化学测菌法检测变形链球菌,建立特定频率下细菌浓度与系统电阻抗值之间的函数关系,依据上述函数关系和测得的电化学阻抗值,分析待检样品内变形链球菌的浓度。结果在1.2KHz固定频率下、变形链球菌浓度为104~109cells/ml时,所测细菌浓度的对数与系统电阻抗值呈良好的线性关系(RITO2=0.995);从进样到获得稳定阻抗数值的时间为6分钟;进样速度采用1ml/h时电化学测菌法的灵敏度最高。结论基于ITO叉指电极微流芯片的电化学测菌法,可为变形链球菌的快速检测提供新的途径。
Objective To investigate the feasibility of the electrochemical method based on ITO electrode in a microfluidic chip for Streptococcus mutans detection.Methods The electrochemical method was used to detect Streptococcus mutans to find the function relationship between the logarithmic value of the cell concentration and the impedance magnitude.Results With the frequency at 1.2kHz and in the concentration range of 104 to 109 cells mL-1,a good linear relationship was found between the impedance magnitude and the logarithmic value of the cell concentration(R2=0.98).From sample injection to stable impedance data collection only used 6 minutes.The sensitivity of the electrochemical method was the best when the inject speed was at 1ml/h.Conclusion The electrochemical method based on ITO electrode in a microfluidic chip might provide a novel approach for rapid Streptococcus mutans detection.
