摘要
目的探讨补肾复方对围绝经期大鼠卵巢超微结构的影响及其作用机制。方法取雌性Wistar大鼠70只,其中4月龄青年鼠(青年组)、18月龄老年鼠(老年组)各10只,灌服生理盐水;11～12月龄围绝经期大鼠50只,分为模型组、对照组及补肾复方高、中、低剂量组各10只,对照组灌服六味地黄汤,补肾复方高、中、低剂量组分别灌服高、中、低剂量补肾复方20mL/kg。灌药4周后取各组大鼠卵巢制成电镜切片观察并拍照。结果模型组大鼠卵泡明显少于青年组,闭锁卵泡增多。应用补肾复方后,生长卵泡增多,与对照组比较,补肾复方各剂量组卵巢的超微结构明显改善,表现在线粒体丰富、有大量的脂滴等,且呈一定的剂量效应关系。结论补肾复方可直接作用于卵巢组织,有类雌激素的治疗作用,能改善卵巢组织的营养状态,有效延缓卵巢组织的萎缩。
Objective To explore the influence of kidney-tonifying recipe on ovarian ultrastructure of perimenopausal rats and its mechanism.Methods A total of 70 Wistar female rats,including four-month-old（youth group） and 18-month-old（old-age group）,with 10 rats in each group,and fifty perimenopausal rats（11-12-month-old）.For the rats in both youth and old-age groups,normal saline was given intragastrically;and perimenopausal rats were divided into model group,control group,and kidney-compound-recipe high-,medium-and low-dose groups,with 10 rats in each group.The rats in control group were given rehmanniae decoction of six ingredients,those in kidney-compound-recipe groups,high-,medium-and low-dose of kidney-tonifying recipe of 20 mL/kg,which contained crude drug of 5.80,3.50 and 1.75 kg/L,was offered to respective group accordingly.After four weeks of medication,the ovarian tissues of rats were collected and inspected under electron microscopy and photographed.Results In the model group,follicles were obviously less than that of the youth group,and atretic follicles increased.After using kidney-compound recipe,the number of growing follicles increased,compared with the control group,the ovarian ultrastructure of rats in kidney-tonifying-recipe groups obviously improved,manifesting abundant mitochondria and a lot of lipid droplets.Conclusion Kidney-tonifying recipe can directly act on ovarian tissue,which has an action similar to estrogen therapy that improves nutritional status and delays atrophy of ovarian tissue.
