摘要
目的探讨炎性因子对骨髓间充质干细胞(BMMSCs)黏附分子表达及对其向心肌梗死区归巢的作用。方法采用全骨髓直接贴壁法获取大鼠原代BMMSCs,体外培养至第3代用于以下实验:①将培养的BMMSCs用浓度为10μg/L的IL-1β刺激24 h,采用流式细胞术检测血管细胞黏附分子-1(VCAM-1)、细胞间黏附分子-l(ICAM-1)的表达率;②开胸结扎雄性SD大鼠左冠状动脉前降支,于心肌梗死后24 h,经尾静脉输注经Brdu标记、10μg/L IL-1β预刺激的BMMSCs;输注BMMSCs 24 h后取心肌组织,做冰冻切片,荧光显微镜下计数组织中的BMMSCs数目。以上实验以设不加IL-1β刺激作空白对照组,另在刺激组加抗VCAM-1单克隆抗体作为实验对照。结果生理状态下,大鼠BMMSCs表面黏附分子ICAM-1表达较高,而VCAM-1表达较低;而IL-1β刺激后,黏附分子VCAM-1表达升高;浓度为10μg/L的IL-1β刺激BMMSCs,输注到大鼠体内,心肌梗死区组织的BMMSCs数目明显高于对照组及IL-1β+抗VCAM-1单克隆抗体组。结论炎性因子IL-1β能够增强大鼠BMMSCs表面VCAM-1的表达,但对大鼠BMMSCs表面ICAM-1的表达无影响;其可以有效促进BMMSCs与血管内皮细胞的黏附,提高BMMSCs向心肌梗死区域迁移,其机制可能与BMMSCs表面VCAM-1表达升高有关。
Objective To observe the effect of adhesion molecules on bone marrow mesenchymal stem cells(BMMSCs) expressing and homing in rats of myocardial infarction.Methods The primary BMMSCs were isolated from rat bone marrow and cultured in vitro and cells at passage and later were used in the following experiments: ① BMMSCs were exposed to 10 μg / L IL-1β for 24 h,analysed the expression of VCAM-1,ICAM-1 on cell surface were performed by flow cytometry;②Male SD rats were subjected to occlusion of left anterior descending artery,5bromodeoxyuridine(Brdu) labeled MSCs,previously treated with 10 μg / L IL-1β were injected by intraveineuse reste 24 h after myocardial infarction;the animals were executed another 24 h later,myocardial tissue was harvested and made into frozen section;the number of BMMSCs was counted under a fluorescent microscope.Results In physiological state,rat BMMSCs express of high level of ICAM-1,while theexpression of VCAM-1 was low;Rat BMMSCs stimulated with 10 μg / L IL-1β,the expression of VCAM-1 increased concentration dependent,while the expression of ICAM-1 did not change markedly;Compared with the control groups,more BMMSCs rested in myocardial tissue of the rat,when the stem cells pretreated with 10 μg / L IL-1β.Conclusion IL-1β can increase the expression of VCAM-1 and can not effect the expression of ICAM-1 on rat BMMSCs;IL-1β can promote BMMSCs adhere to endothelial cells,promote BMMSCs migrate to the myocardial tissue.The mechanism may be concerned with increase of VCAM-1.
