摘要
目的:建立测定大鼠肾组织中硫酸依替米星含量的方法。方法:采用反相高效液相色谱柱前衍生化法,样品处理时加入异丙醇沉淀蛋白,以邻苯二甲醛进行衍生化。色谱柱为Platisil ODS C18,流动相为以水-冰醋酸-甲醇(20:5:75)制备的0.02mol.L-1的庚烷磺酸钠溶液,检测波长为330nm。检测大鼠单次肌肉注射硫酸依替米星2mg.kg-1后第1、2、4天以及连续5d腹腔注射硫酸依替米星60、100mg.kg-1.d-1后肾组织中的药物浓度。结果:肾组织匀浆中硫酸依替米星检测浓度线性范围为1.25~60μg.mL-1(r=0.9973)。肌肉注射硫酸依替米星第1、2、4天肾组织中的药物含量分别为(32.88±3.21)、(37.67±4.93)、(21.63±4.91)μg.g-1;连续腹腔注射60、100mg.kg-1.d-1后肾组织中的药物含量分别为(241.4±158.5)、(386.4±98.3)μg.g-1。结论:所建立的方法操作简单、结果可靠,可用于肾组织中硫酸依替米星的含量测定。
ABSTRACT OBJECTIVE: To establish the method for the content determination of etimicin sulfate in renal tissue of rats. METH- ODS: Pre-column derivation RP-HPLC method was adopted. Protein was precipitated by isopropanol, and the sample was derivative with o-phthalaldehyde. The separation was performed on a Platisil ODS C18 column with 0.02 mol.L-1 sodium heptanesulfonate pre- pared by a mixture consisting of water-acetic acid-methanol (20:5:75) as mobile phase at the UV detection wavelength of 330 nm. After the delivery of etimicin sulfate by a single-dosage intramuscular injection at the dosage of 2 mg.kg-1, the drug concentration was determined at 1 d, 2 d, 4 d. After the delivery of etimicin sulfate (60, 100 mg.kg-1.d-1) by peritoneal injection for 5 consecu- tive days, the drug concentration was also determined. RESULTS: The linear range of etimicin sulfate in renal homogenate was 1.25-60 μg.mL-1 (r=0.997 3). The concentration of etimicin sulfate in kidneys at 1 d, 2 d, 4 d after intramuscular injection were (32.88 ± 3.21) μg.g-1, (37.67 ±4.93) μg.g-1 and (21.63 ± 4.91) llg.g-1, respectively. The concentration of etimicin sulfate in kidneys after peritoneal injection at dosage of 60, 100 mg.kg-X.d-1 were (241.4 ± 158.5) μg.g-1 and (386.4± 98.3)μg.g-1, respectively. CONCLUSION: The method is simple and relibale, which is suitable for the content determination of etimicin sulfate in kidney.
